摘要:Syndromes,seen from the objective phenomenon,are the assemblage of the macro information of human being(symptoms and signs).The author analyzed the formation procedure of syndrome system from the human cognitive process,and drew the conclusion that the process of syndrome system formation is that of linking the different symptoms and signs together according to some specific features.This link process starts from the concordance of time and is complemented by the concordance of the identity of forming factors,parts and nature,and is finished by the concordance of impressive factors(especially the therapeutic factors).Therefore,the therapeutic factor is a vital evidence for the foundation of syndrome system.
摘要:Objective To study the effect of Cordyceps mycelia extract(CME) on injury and phenotype shift of liver sinusoidal endothelial cells(SECs) in rats with dimethylnitrosamine(DMN)-induced liver cirrhosis.Methods The rat liver fibrosis model was established by peritoneal injection of DMN(at a dose of 10 mg·kg-1,3 times a week,for 4 wks).The dynamic changes of liver cirrhosis were observed at different time points(3 d,2 w,4 w).Liver ultrastructure was observed by electron microscope.The expressions of CD44 and von Willebrand factor(vWF) were examined by immunohistochemistry.The serum hyaluronic acid(HA) content was measured by radioimmunoassay.On the meantime CME were taken orally by 0.74 g·kg-1·d-1.Results The numbers of CD44 positive staining SECs in the 3 d model rats were obviously less than those of normal rats(P<0.05);vWF positive staining increased;the numbers of fenestrae rapidly decreased,but serum HA content rapidly increased(P<0.05).The defenestration and formation of basement membrane of SECs were observed in the 4 wk model rats.In the CME-treated 3 d and 2 wk rats,the defenestration and formation of basement membrane of SECs were alleviated.In the CME-treated 2 and 4 wk rats,serum HA content decreased(P<0.05),CD44 expression increased,vWF positive staining decreased,the numbers of fenestrae increased,and formation of basement membrane decreased.Conclusions CME is able to decrease the injury and phenotype shift of SECs,inhibit the formation of capillarization in rats with the DMN-induced liver cirrhosis.
摘要:Objective To investigate the effects of genistein(GEN) on ovary genes differential expression in CDDP induced premature ovarian failure(POF) mice with syndrome of kidney and liver yin asthenia,and its mechanism.Methods POF mouse model with syndrome of kidney and liver yin asthenia was established by chemotherapy injury according to literature.The ovary total-RNAs of GEN group and normal control group were extracted and were reversely transcribed with Cy3,Cy5 fluorescent labeling to obtain cDNA probes that were hybridized with cDNA microarray chips.The results from the laser scanner were made image analysis,standardization,ratio analysis and cluster analysis.Results Sixty genes expressed were screened out differentially;22 were up-regulated and 38 were down-regulated.These Genes were classified into three groups,which were cell composition,molecule function and biology process.Pathway analysis indicated that GEN participated in the process of ribosome gene expression and controlled the GnRH and MAPK gene expression mainly.Conclusiosn There are many different gene expression pattern in POF mouse with syndrome of kidney and liver yin asthenia.GEN may remove functional restriction of RNA partly and up-regulated ATP metabolism related gene expression,which will strengthen energy metabolism.

摘要:Objective To observe dynamically the influence of salvianolic acid B(SalB)on secretive function of vascular endothelial cells in mice with cerebral ischemia reperfusion.Methods NIH mice were randomly divided into sham-operated group,model group,SalB-treated group and Nimodipine-collated group.The model with cerebral ischemia-reperfusion was established.After ischemia for 30 min followed by 0 min,30 min,60 min of reperfusion,Nitrogen monoxidum(NO) content from brain tissue was determined,and the contents of endothelin(ET),tissue-type plasminogen activator(t-PA),plasminogen activator inhibitor-1(PAI-1) and von Wilebrand factor(vWF) in blood plasm were measured respectively.Results At diefferent time after cerebral ischemia reperfusion injury,contents of ET,NO,PAI-1 and vWF improved remarkably,whereas t-PA content decreased in the model group compared with the sham-operated group.However,SalB regulated secretive function of endothelial cells,especially at 60 min after reperfusion.It decreased markedly contents of ET,NO,PAI-1 and vWF(P﹤0.01 or P﹤0.05),and improved content of t-PA(P﹤0.01).Conclusion SalB can protect cerebral ischemia reperfusion from injury,which may contribute to its action on regulating the balance among dilatation,contraction,procoagulant and anticoagulant substances secreted by vascular endothelial cells.

摘要:Objective To observe the effect of Weiyanyin on the expression of Cyclooxygenase-2(COX-2)mRNA gastric mucosa of rats with experimental bile reflux gastritis.Methods The model of experimental bile reflux gastritis was eshtablished by feeding reflux liquid.Domperidone was taken as control.The effect of Weiyanyin on pathological histology was observed and the expression of COX-2mRNA in gastric mucosa was detected by method of RT-PCR.Results The gastric mucosa of rats in the model group was damaged,amotic,and a lot of inflammatory cells could be seen.Most of rats showed glandular organ hyperplasy and intestinal metaplasia.At the same time the expression of COX-2mRNA were increased(P<0.01).Compared with the model group,Weiyanyin could improve pathological histology of experimental bile reflux gastritis rats,and decrease the expression of COX-2mRNA(P<0.05 or P<0.01).Conclusion Weiyanyin could decrease the expression of COX-2mRNA to prevent mucosal precancerous change such as Intestinal metaplasia,atypical hyperplasia and so on.

摘要:Objective To investigate the effect of gradient changes of cold pathogen on the ultra-structure of lung in rats.Method 56 Wistar rats of SPF grade were divided into normal temperature group,absolute temperature groups(including 3 groups of 10 ℃,0 ℃ and-10 ℃) and temperature difference groups(including 3 groups of 2010 ℃,200 ℃ and 20-10 ℃) by using random digits table,with 8 rats in each group.The ultra-structure of lung in rats was observed under the transmission electron microscope.Results There were different degrees of changes in the ultra-structure of lung in groups with abnormal environmental temperatures.With the decrease of the temperature and the increase of the temperature difference,the pathological changes of lung aggravated.There were no marked changes in alveolar epithelial cells of rats in the group of 10℃ and 0℃,while there were obvious changes in the group of-10℃ and all of the temperature difference groups.The pathological changes of ultra-structure of lung were more severe in the absolute temperature groups than those in the temperature difference groups.Conclusion The effects of gradient changes of cold pathogen on the ultra-structure of lung in rats are different.The cold pathogen is related not only with the absolute temperature but also with the temperature difference closely.

摘要:Objective To observe the effect of Jiaweiqinglin Pills(JP) on antioxygenation in aged rats to investigate the its antidotal mechanism.Method After adaptability feeding for one week,120 healthy Wistar rats were randomly divided into 6 groups,which were 4-month control group(4C group),14-month control group(14C group),14-month JP group(14JP group),24-month control group(24C group),24-month JP group(24JP group) and(14-24)-month JP group(14-24JP group),with 10 males and 10 females in each group.All rats were put to death after the last time medication and fasting for 12 hours.SOD and MDA in serum and in the tissues of heart,brain and liver were detected,and glutathione peroxidase(GSH-PX) activity in serum was measured.Results Vitality of SOD in serum and liver tissue of 14C group is lower than those of 4C group(P<0.01)and 14JP group(P<0.01).But it was not obvious in the heart and kidney tissues.Contents of MDA in serum and in the heart and liver tissue of 14C group were higher than those of 4C group(P<0.01)and 14JP group(P<0.05).Vitality of GSH-PX in serum of 14C group is lower than it of 4C group(P<0.01)and 14JL group(P<0.01).Vitality of SOD in serum,heart liver and kidney tissue of 24C group is lower than it of 24JL group(P<0.01)and 14-24JL group(P<0.01).To the best obvious is the vitality of SOD in serum..Vitality of GSH-PX in serum of 24C group was lower than that of 24JP group(P<0.01)and 14-24JP group(P<0.01).Contents of MDA in serum and in the heart,liver and kidney tissues of 24C group were higher than those of 24JP group(P<0.01)and 14-24JP group(P<0.05),especially in the serum.The MDA of kidney of 24JP was lower than that of 14-24JP.Conclusion Jiaweiqinglin Pills may enhance the antioxidation mechanism of aged rats.
摘要:Objective To observe the effect of β-aescinate administered on changes of edema after intracerebral hemorrhage in rats and to explore the mechanism.Methods Intracerebral hemorrhage rat models were established by stereotaxically injecting autologous blood at caudate nucleus.Rats were randomly divided into two groups: test group and control group.After operation,β-aescinate was injected by caudal vein in different time(1 h,3 h,6 h and 24 h) to test group(2.5 mg/kg·d) and sodium chloride was injected to control group in corresponding time.Brain water content was investigated and the expression of TNF-α,IL-6 and MMP-9 was measured in brain tissue.Results Water content in the perihematoma tissue in the test group was lower than that in the control group(P<0.05).However,there was no statistic significance between subgroups of treatment(P>0.05).Compared with the control group,the edema of neuron and stellate cell were lower in test group;the expression of TNF-α in brain tissue were obviously lower(P<0.05) in test group.At the same time,the expression of IL-6 in brain tissue had no significant difference between the test group and the control group(P>0.05),but MMP-9 in the control group was higher than that in the test group(P<0.05).Conclusion β-aescinate has an evident protective effect on resisting edema after intracerebral hemorrhage in rats,which may be related to anti-inflammatory.
摘要:Objective To investigate the effects of Naoshuning,a TCM formula for activating blood circulation and alleviating water retention,on the protein expression of aquaporin4(AQP4) in experimental injured brain tissue of rats and its mechanism.Methods The SD rats were randomly divided into the sham operation group(n=88),the traumatic brain injury(TBI) group(n=88) and the Naoshuning group(n=88).Changes of brain water content,permeability of blood-brain barrier(BBB) were determined at 6 h,1 d,3 d and 5 d after TBI.Immunohistochemical technique was used to investigate the protein expression of AQP4.Results The brain water content,permeability of BBB and AQP4 protein expression of TBI group were obviously higher than those of the sham operation group(P<0.001).The electron microscope result indicated that BBB was evidently damaged 1day after TBI,with vascular endothelial cell tight junction damaged,basement membrane broken and astrocyte end feet process swelled obviously.In Naoshuning treatment group brain tissue water content,EB content and the expression of AQP4 were obviously lower than those of the model group at each time point(P<0.05).The ultramicrostructure changes could be relieved by Naoshuning.Conclusion The principle of activating blood circulation and alleviating water retention showed effects in reducing cerebral edema and protecting BBB,which may be related with inhibiting the epression of AQP4 protein.
摘要:Objective To establish a HPLC method for the quantitative determination of four alkaloids,i.e.2-hydroxy-1-methoxyaporphine,pronuciferine,nuciferine and roemerine,in Nelumbo nucifera.Methods The determination was carried out with Hypersil C18 column(250 mm × 4.6 mm,5 μm),using acetonitrile-water(0.1% triethylamine) as mobile phase with gradient elution,at a flow rate of 1.0 mL·min-1 and detected at the wavelength 270 nm,column temperature 35 ℃.Results The linear ranges of 2-hydroxy-1-methoxyaporphine,pronuciferine,nuciferine and roemerine were 0.109 6~0.657 6 μg(r=0.999 5),0.021 0~0.126 0 μg(r=0.999 5),0.103 0~0.618 0 μg(r=0.999 8) and 0.085 6~0.513 6 μg(r=0.999 5),with the average recoveries(n=6) of 99.5%,100.5%,97.5% and 100.1% respectively.The contents of four components in different samples of Nelumbo nucifera were: 2-hydroxy-1-methoxyaporphine 0.01%0.076%,pronuciferine 0.002%0.120%,nuciferine 0.008%0.249%,roemerine 0.008%0.067.Conclusion The contents of 4 components in 30 market samples varied notably,especially for pronuciferine and nuciferine,and should be taken into the quality control of Nelumbo nucifera.
摘要:Objective To observe the effect of acupotomy on pain threshold and monoamine neurotransmitters in spinal cord and the hypothalamus of rats with the transverse process syndrome of third lumbar vertebrae.Methods Twenty-eight rats were randomly divided into four groups: the normal control group,the model group,the electroacupuncture(EA) group and the acupotomy group.The last three groups were modeled by an operation.The EA group and the acupotomy group were treated respectively.The changes of rats’ pain threshold were observed with thermal radiation-resistant method.28 days after modeled,monoamine neurotransmitter contents of hypothalamus and spinal cord were detected with RP-HPLC.Results The value of pain threshold was significantly lower in the model group than in the normal group(P<0.05) and was higher in the EA group and the acupotomy group than in the model group with no significant differences compared with the normal group(P>0.05).The contents of 5-HIAA of the spinal cord in the EA group the acupotomy group were significantly higher than that in the model group(P<0.05),but there was no significant difference compared with the normal group(P>0.05).The content of DOPAC in the hypothalamus in the EA group were more than that in the other three groups(P<0.05).The content of DOPAC in the spinal cord in the acupotomy group was significantly lower than that in the other three groups(P<0.05).The content of 5-HT,NE and DA in both the spinal cord and the hypothalamus were no significant differences among four groups(P>0.05).Conclusion Acupotomy’s analgesic effects on the rat model may through the promotion of metabolism and liberation of some monoamine neurotransmitter.Its’ effect may be better than electroacupuncture in this respect.

摘要:Objective To discuss the protective mechanism of electroacupuncture(EA) in the acute spinal cord injury(SCI).Methods Rats were divided into five groups stochasticly: blank group,sham operated group,model group,drug group and EA group.The model rats were made a spinal cord contusion by using a modified Allen’s method.After SCI the EA group was treated by "mingmen"(DU4) and "dazhui"(DU14) points;the drug group was treated by methylprednisolone(MP).Six hours after SCI,the rats were put to death.The calcium ion content in serum was detected and the differentially expressed genes were investigated by bioinformatics technique.Results The calcium ion content in serum of the EA Group decreased significantly as compared with those in the model group(P<0.05).There were 266 differentially expressed genes in the model group and 181 differentially expressed genes in the EA group.Conclusion EA had protective effect on the SCI through depressing the calcium ion content.There were lots of differentially expressed genes in SCI rats.These differentially expressed genes are relative with protective effect,growth and regeneration of the nerve tissues.

摘要:objective To observe the curative effect and safety of treating RA(syndrome of qi deficiency and dampness-heat) by using Sishenjian.Method Fifty patients were randomly assigned to two groups: the treatment group(Sishenjian) and the control group(Shirebi).The curative effect and the safety were observed and compared between the two groups.Results The curative effect of the "Sishenjian" group was superior to that of the control group with a statistical significance.The total effective rates in the "Sishenjian" group and in the control group were 92.0% and 65.2% respectively;and the excellent rates were 32.0% and 8.7% respectively.Statistical results indicated that "Sishenjian" remarkably lowered the symptom-marks of RA and alleviated the discomforts caused by RA.Patients of the Sishenjian group didn’t show unhealthy reactions,so it’s safety and reliability.Conclusion "Sishenjian" is mainly used for treating RA with syndrome of qi deficiency and dampness-heat,especially for patients with swollen and painful joints which skin-temperature is high.The clinical effect of "Sishenjian" is good and with higher safety.

摘要:Objective To investigate the reliability,validity and responsiveness of Chinese version of National Institutes of Health Stroke Scale(NIHSS)in big sample patients of multi-center with stroke in China.Methods The reliability was evaluated in 537 inpatients by using Cronbach’s α coefficient,Guttman Split-half and Spearson correlation coefficient.The content validity of NIHSS was studied with the method of subjective reasoning and judging.The construct validity was assessed with factor analysis.Criterion validity of individual domains was assessed by comparing the linear association of the domain score with the score of these scales.Domain responsiveness was assessed with Paired Ttest.Results All scales of 14 d and 21 d showed better internal consistent.Most domains except gaze and ataxia domain demonstrated excellent reliability in test retest(Spearson correlation coefficient ranged 0.5-1).All 12 domains demonstrated excellent constract validity(accumulative dedication rate ranged from 88.6 to 90.17%). NIHSS total score and every item of the most domains in different time points except gaze domain of 7d were responsive.NIHSS showed high correlation with Glasgow Outcome Scale(GOS)and Barthel Index(BI).Conclusion The Chinese version of NIHSS is a reliable,valid and responsive scale and is suitable for assessing the therapeutic effect of Chinese medicine on stroke at acute phase.
