      摘要:Objective To build a rat model of Qi deficiency and blood stasis syndrome with cerebral ischemic stroke and give an evaluation on the model.Method Middle cerebral artery occlusion(MCAO) model was made after 3 weeks’ continual exhaustive swimming.Data of exterior signs,tongue image,hemorheology and morphology were collected to establish a evaluative system of the model.Result The model rats showed a slower weight gain,postponed intake,darker tongue and longer sublingual veins in the course of continual exhaustive swimming,which reflects a state of Qi deficiency and blood stasis.And the model rats were inactive,weak,hemiplegic,and showed a dark purple tongue and longer blue sublingual veins after MCAO,which were the hints of Qi deficiency and blood stasis syndrome in cerebral ischemia.There were significant increases in blood viscosity,plasma fibrinogen content and platelet aggregation rate in the model group.And brain tissues morphology showed ischemic changes in model rats.Conclusion Continual exhausting swimming followed by MCAO could be a method to make a rat model of Qi deficiency and blood stasis syndrome with cerebral ischemic stroke,which could be evaluated by integrated criteria including exterior signs,tongue image,hemorheology and morphology,etc.

      摘要:Objective To inquire into the cerebral gene changes in mice with kidney-yang deficiency caused by overwork and intemperance in sexual activity following the treatment of Jinguishenqi Pills(JP) or Yougui Pills(YP).Methods Forty-five male mice were divided into 4 groups,with 10 in the control group,15 in the model group,10 in the JP group and 10 in the YP group.Every mouse in the control and model group was given distill water 0.5 mL per day,and every mouse in the JP and YP group were given 0.5 mL suspension of JP or YP per day.The male mice in the model,JP and YP group lived in the same cage with female mice at the ratio of 1∶ 6(male:female),and swam 30~40 min everyday for 4 weeks to result in kidney-yang deficiency caused by overwork and intemperance in sexual activity.The brain gene expression of the mice were detected with mouse brain gene chip of 36K mouse genome array made by CapitalBio Corp.Beijing,China.Results Compared with the control group,186 differential expression genes were screened in the model group,including 150 up-regulated genes and 36 down-genes;299 differential expression genes were screened in the JP group,including 114 up-regulated genes and 185 down-regulated genes;155 differential expression genes were screened in the JP group,including 64 up-regulated genes and 91 down-regulated genes.There were 24 up-regulated genes both in the JP and YP group,accounting for 21.05 percent and 37.5 percent in each group respectively,which were mainly the genes related to neurotransmissions/signal transduction,transcription/translation,cellular cycle,hormone and metabolism.The down-regulated genes both in the GP and YP group were 35,accounting for 18.92 percent and 38.46 percent in each group respectively,chiefly including the genes associated with inflammation/immunization,neurotransmissions/signal transduction,transcription/translation,metabolism and limiting velocity enzyme of dopamine.Compared with the control group and model group respectively,there were 23 genes up-regulating in the model group while down-regulating in the treatment group,mainly involving the genes related to inflammation/immunization and neurotransmissions/signal transduction.There were six genes down-regulating in the model group,while up-regulating in the treatment group,mainly including the related genes of cellular cycle and structure,neurotransmissions/signal transduction and transcription.Conclusion JP and YP may down-regulate the expressions of the genes related to hormone and melanin-concentrating hormone(MCH) which are up-regulated in mice with kidney-yang deficiency.This study can inquire into the effect mechanism of the two drugs and their differences in genetic level.

      摘要:Objective To study the relationships of pre-menstrual syndrome(PMS) and expression of GABAA receptor β2 subunit mRNA in hippocampus.Method The expression of GABAA receptor β2 subunit mRNA in rat hippocampus was detected by semi-quantitative RT-PCR between normal group,model group and Baixiangdan Capsules group.Result Compared with the normal group,the expression of GABAA receptor β2 subunit mRNA was significantly high in hippocampus of the model group,and Baixiang dan Capsules decreased the high expression of it.Conclusion The pathogenesis of PMS with the syndrome of reversed liver qi is concerned with GABAA receptor subunit β2 in hippocampus.

      摘要:Objective To establish an animal model with blood stasis syndrome by combination of cerebral ischemia and cool irritation,and compare it with two existing animal models with blood stasis syndrome in order to select an optimal exosymptom model of blood-stasis syndrome.Methods Three rat models of blood-stasis syndrome were selected,including middle cerebral artery occlusion model(MACO),cool irritation model(CI) and combination of middle cerebral artery occlusion and cool irritation model(MACO&CI).The exosymptoms of blood stasis syndrome were observed at different time points after cerebral ischemia(on 1st,3rd,5th day and 7th day).Ethological and haemorheological detection were performed.Results Compared with the normal group,tongue color index values(TCIV) of rats were high 3days after cerebral ischemia in the MCAO,CI and MACO&CI groups,with significant differences(P<0.05);only high in the MACO&CI group 5 days after cerebral ischemia(P<0.05).TCIV of the MACO&CI group was the highest(60.12±5.154) 3 days after cerebral ischemia in all time points.The whole blood viscosity,reduced viscosity,plasma viscosity,hematocrit,index of erythrocyte aggregation and deformity index of RBC of rats in the MCAO,CI and MACO&CI groups were higher than those in the normal group,with significant differences at some time points(P<0.05).Among all groups and all time points,in the MACO&CI group 3 days after cerebral ischemia the blood viscosity,hematocrit,plasma viscosity and index of erythrocyte aggregation were the highest and the deformity index of RBC was the lowest.Conclusions The animal model of combination of cerebral ischemia and cold stimulation was one rational exosyndrome model of blood stasis syndrome.

      摘要:objective To investigate the effects of Qiangji Tablets on learning-memory impairment mice induced by scopolamine and its possible mechanism.Methods Qiangji Tablets was orally administered for 14 days.Mice learning-memory deficit was induced by scopolamine intraperitoneal injection 30mins before Morris Water Maze and Step-down avoidance behavior test,then the brain of mice was dissected for ChAT and AChE activity assay.Results Qiangji Tablets blocked learning-memory impairment induced by scopolamine,increased the activity of ChAT and decreased the activity of AChE in the mice brain with dose-effect relationship in some degree.Conclusion Qiangji Tablets improves mice learning-memory ability and enhances mice acetylcholinergic system.The results indicate that Qiangji Tablets may have a promising application in treatment of dementia diseases such as Alzheimer’s Disease.

      摘要:Objective To study the effect of Ningxinhongqi Capsules(NC) on microstructure of aged female rats’ ovarian granular cells.Methods Fifty natural aged 14-month-old SD rats were equally randomized as five groups and supplied with phytoestrogen-free diet for 8 weeks:menopausal model group with normal diet,positive control group with ethylene estranol(EE) at 0.18 mg·kg-1 per day,medicine group with heavy dose intake of NC at 0.648 g·kg-1 per day,middle dose intake of NC at 0.324 g·kg-1 per day,and small dose intake of NC at 0.162 g·kg-1 per day.Ten natural aged 4-month-old SD rats were used as a healthy control group and supplied with phytoestrogen-free food.Then the ovarian granular cells were taken for further observation under the electron microscope and morphometric analysis by using image analysis software.Results Compared with the model group,the indexes including numerical density,perimeter density,area density and average area of mitochondria and smooth endoplasmic reticulum in the medicine group increased significantly(P<0.05).Conclusion Ningxinhongqi Capsules can improve the situation of mitochondria and smooth endoplasmic reticulum both in number and configuration to delay ovarian aging.

      摘要:Objective To study the pathologic changes of liver in type Ⅱ diabetes model KKAy mice and the protective effect of puerarin on liver injury in KKAy mice,and to find a new strategy of diabetes-protecting the structure and function of liver.Methods Male KKAy mice,9~11weeks,were divided into model group(n=20) and puerarin group(n=20) randomly.Male C57BL/J mice,9~11weeks,were control group(n=20).Body weight was measured at 16,20,24,28 weeks of age and plasma-glucose was measured at 24,28 weeks.Mice were sacrificed at 24 and 28 weeks of age respectively,and lipids,alanine aminotransferase(ALT) and aspartate aminotransferase(AST) were measured.Hepatic tissue was morphologically observed by light microscope.Results 1.The body weight of mice was controlled by puerarin before 24 weeks of age(P<0.05).2.Lipids,ALT and AST of KKAy mice in the puerarin group were lower than that in the model group at 28 weeks of age(P<0.05).3.At 28 weeks of age,the structure of liver was damaged,and fatty degeneration in the hepatocyte was significant.Conclusion Using puerarin in early period can protect the hepatic structure and function in KKAy mice.

      摘要:Objective To explore the effect of brain injuries of Parkinson’s disease on proliferation of neural stem cells in brain and role of ginkgo biloba extract.Method C57BL/6 mice were randomly divided into normal group,model group and treatment group.Parkinson’s disease model was made by i.p.1 - methyl,4 - phenyl - 1,2,3,6 tetrahydropyridine.Proliferation and migration of neural stem cells in dentate gyrus in every group was observed by immunohistochemistry and double labeled immunohistochemistry technique.Result On 28th day,the number of the PCNA positive cells was increased significantly in model group(MG) and ginkgo biloba extract group(EGBG),but the increasing in MG was more than that in EGBG.The number of the nestin positive cells in dentate gyrus was increased in MG and EGBG on 7th and 28th day,and the increasing in EGBG was the most obvious on 7th day.The number of the PCNA/Nestin double positive cells in dentate gyrus was increased in MG and EGBG on 7th and 28th day,and the increasing in EGBG was the most on 28th day.Conclusion The brain injuries of Parkinson’s disease may induce proliferation of neural stem cells in brain.Ginkgo biloba extract could promote proliferation and migration of neural stem cells in brain of Parkinson’s disease model mice.

      摘要:Objective To evaluate curative effect of TCM objectively on children’s respiratory syncytial virus pneumonia with "wind and heat blocking the lung syndrome" and "phlegm-heat obstructing the lung syndrome".Methods Single blind,multi-center,stratified randomization and equal control methods were used on 297 cases of RSV pneumonia with "wind and heat blocking the lung syndrome" and "phlegm-heat obstructing the lung syndrome" in five centers hospitalization who were observed and treated by Chinese medicine or western medicine.In the control group,the patients were treated by Ribavilin Injection and Compound guaiacol potassium sulfonale Oral Solution;In the test group,Qingkailing Injection and Ertongqingfei Oral Solution were used for patients with "wind and heat blocking the lung syndrome",and Qingkailing Injection and Xiaoerkechuanling Oral Solution for "phlegm-heat obstructing the lung syndrome".The time of therapy was 10 days for each group.Reults The score of primary symptoms at post-treatment and the score diminishment between pre-treatment and post-treatment in the test group,especially in the group with the "phlegm-heat obstructing the lung syndrome",were better than those in the control group(P<0.01).The symptom recovery in fever,cough,sputum and auscultation on the lung of the test group were much earlier than that of the controlled group(P<0.01).The total effective rates evaluated by disease and score’s diminution of primary symptoms were 90.54% and 87.84% in the test group,and were 81.88% and 65.77% in the control group,both with significant differences between two gruops(P<0.05,P<0.01).At the same points for the "wind and heat blocking the lung syndrome",the total effective rates were 88.89% and 91.67% in the test group,and were 80.62% and 64.29% in the control group,both with significant differences between two gruops(P<0.05,P<0.01);and for the "wind and heat blocking the lung syndrome",they were 95.0% and 77.50% in the test group and 84.31% and 68.63% in the control group,both without significant differences between two gruops(P>0.05).Conclusion TCM has marked clinical curative effects on children’4s respirator syncytial virus pneumonia,especially on that with the "phlegm-heat obstructing the lung syndrome".

      摘要:Objective To study the effect of integrated traditional Chinese and western medicine on residual renal calculus after different surgical methods in patients having been treated with extracorporeal shock wave lithotripsy(ESWL).Methods 100 patients with residual renal calculus were divided into A,B and C groups after ESWL.The patients in A group were treated with western medicine,the patients in B group were treated with integrated traditional Chinese and western lithagogue decoction,and the patients in C group accepted integrated traditional Chinese and western lithagogue decoction and Niaolutong(compound tablets).Two weeks were considered to be one course of treatment in each group and 4 courses were followed up.Results The cure rate of renal calculus in A,B and C groups were 69.70%,93.94% and 94.12% respectively;the cure rate in A group was significantly lower than that in B or C group(P<0.05).Conclusion Integrated traditional Chinese and western medicine can efficiently increase the cure rate of residual renal calculus,accelerate the elimination of calculus and shorten the treatment time.

      摘要:Objective To study the protective effect of Shenfu Injection(SFI) on myocardial cells in patients undergoing coronary artery bypass grafting(CABG).Methods Sixty patients undergoing coronary artery bypass grafting during Dec.2005 to Dec.2006 were randomly divided into SFI group(30 cases) and control group(30 cases).Cold crystal cardioplegic solution was infused through ascending aorta at initial time,20 min after blood flow being blocked,the cardioplegic solution containing blood was infused every 20~30 min in the control group,in the same condition SFI 20 ml/L was infused in combination with the cardioplegic solution containing blood in the SFI group.Results The serum levels of troponinI(cTnI) and the hemodynamics were observed in these patients at different time points.There were no significant differences between two groups in the level of cTnI and hemodynamics before operation.There were significant differences between two groups in the level of cTnI and hemodynamics after aortic declamping(P<0.05).Conclusion Shenfu Injection can attenuate the degree of myocardial damage in patients undergoing CABG.

      摘要:Objective To study the therapeutic efficacy of manipulative therapy in treating wind-cold type of common cold.Methods 208 cases of wind-cold type of common cold were divided into two groups randomly.The patients in the manipulation group(105 cases) were treated by manipulative therapy,and the patients in the drug group(103 cases) were treated by Contac’NT.After 3 days treatment,statistical analysis was made to analyze the treatment outcome of two groups.Results The total effective rate was 87.6% in the manipulation group(recovery,20cases;significant effect,53cases;effectiveness,19cases;inefficiency,13cases) and was 89.3% in the drug group(recovery,19cases;significant effect,51cases;effectiveness,22cases;inefficiency,11cases).There was no statistical significances in the therapeutic efficacy between the two groups(P>0.05),but there was statistical significances in the symptom scores between the two groups(P<0.05).Conclusion Manipulative therapy is effective in treating the wind-cold type of common cold,and is easy to operate without side effects.

      摘要:Objective To investigate the effects and neuro-endocrine mechanism of mild needle therapy of mongolian medicine on fatigue rats.Methods Male SD rats were evenly randomized into control,model and mild needle groups.The model was established by exhausted swim.Mild needle therapy of mongolian medicine was applied to Dinghui and Xinxue for 15 min,once every 2 days.On the 21st day of the model establishment,the exhausted swim time,the immobility time in tail suspension test were measured.24 h after the last swim,the rats were decapitated.The norepinephrine(NE),dopamine(DA),dihydroxy-phenyl acetic acid(DOPAC),homovanillic acid(HVA),5-hydroxytryptamine(5-HT),5-hydroxyindoleacetic acid(5-HIAA) in hypothalamus were detected by HPLC-ECD method.The malondialdehyde(MDA),glutathione(GSH),glutathione peroxidase(GSH-PX) and superoxide dismutase(SOD) in serum and testis were measured.The corticosterone and testosterone content in serum were detected by ELISA.The testis index and adrenal gland index were calculated.Results Mild needle therapy significantly prolonged the exhausted swim time(P<0.05),and reduced the immobility time in tail suspension test(P<0.05).It also lowered the 5-HIAA and HVA content in hypothalamus(P<0.05,0.05).It significantly increased GSH in serum and testitis(P<0.05,0.05) and increased SOD in testitis(P<0.05),and significantly reduced corticosterone(P<0.01) and improved T/C(P<0.01) in serum.There were no significant differences among 3 groups in body weight,testis index and adrenal gland index(P>0.05).Conclusion Mild needle therapy of mongolian medicine can increase the monoamine content in rat brain,and improve the activity of antioxidant substances in the body.It may also reduce corticosterone in serum,improve T/C,which suggest it may regulate the hyperactivity of HPA axis.So they may underlie the action of mild needle therapy in raising the exercise ability and improving fatigue.
