摘要:The content and methods of macroscopic physical signs collected from rats were discussed from three aspects,including the information obtained by TCM four diagnostic methods based on the characterization of the appearance such as body,activities,skin and fur,eyes,tail,paw,nail,lips,urine and stool;the changes of value and imagines obtained by using some instruments such as body temperature,electrocardiogram and electrophysiology reflecting the macroscopic physical signs;the evaluation of the macroscopic physical signs based on methodology including methods of quantification and semi-quantification.The train of thought and methods were found out to review the macroscopic physical signs of animal model through the observation and collection of these signs.

摘要:Combined the achievemetns of prophase researches,it was thought that,based on the analysis on the relationship between aryl-alkylated amine N-acetyltransferase(AANAT)-melatonin-pineal gland and TCM regulation theory of five zang-viscera,pineal gland was a medium in the relation between five zang-viscera regulation and five seasons,and melatonin might be a set point in the correspondence between five zang-viscera and five seasons.AANAT,as a main rate-limiting enzyme of melatonin synthesis,had seasonal changes in its activity,which determined the rate and rhythm of melatonin synthesis and played a very important role in melatonin concentration.AANAT-melatonin-pineal gland was thought as the breakthrough point in the research of TCM regulation theory of five zang-viscera.

摘要:It is thought that the reducing analysis method can not be the core direction of discipline research through the analysis on the basic study of science of herbal formula at present.The changes of micro-index can not replace the changes of macro-function and can not be taken as the modernization standard,while real modernization of micro-index and macro-function should be confirm and complement each other.The kernel content of science of herbal formula should be focused on the research of therapies besides the laws of formula design and correlation between formula and syndrome.The research method starts from traditional theory and clinical practice and takes the course of original theory.The science of herbal formula depends on the theoretical innovation for its long-term development,so it is not only a tool in the research of TCM patent medicine.

摘要:Objective To explore the influence of Tongsai Granules on inflammatory cells and lung function in rat model of chronic obstructive pulmonary disease(COPD)in acute exacerbation stage.Method Wistar rats(n=70)were randomly divided into 7 groups,including the control group(group 1),model group of COPD in stable stage(group 2),model group of COPD in acute exacerbation stage(group 3),high-dose Tongsai Granules group(group 4),low-dose Tongsai Granules group(group 5),aminophylline group(group 6)and Tingbei Capsules group(group 7,each n=10).The model of COPD in acute exacerbation stage was established by repeatedly bacterial infection of respiratory tract.Group 1,2 and 3 were given normal saline solution intragastrically twice a day,and group 4,5,6 and 7 were given intragastrically Tongsai Granules(5 g/kg/d and 2.5 g/kg/d),aminophylline(2.3 mg/kg/d),and Tingbei Capsules(0.476 g/kg/d)respectively twice a day.The medicine administration time was one week.The influences of Tongsai Granules on inflammatory cells in peripheral blood(PB)and bronchoalveolar lavage fluid(BALF),and lung function were observed.Result In group 3 the counts of white blood cells(WBC)and neutrophil(N)in PB were higher than those in group 1 and 2(P<0.01,P<0.05),and those were higher in group 4 than those in group 6(P<0.05).The count of N in PB was higher in group 4 than that in group 7(P<0.05).In group 4 the counts of WBC and N in PB were lower than those in group 6(P<0.05).In group 2 the counts of WBC and N in BALF were higher than those in group 1(P<0.05,P<0.01),and lower than those in group 3(P<0.01).In group 4 and 5 the counts of WBC and N in BALF were significantly lower than those in group 3(P<0.05,P<0.01)and group 7(P<0.05,P<0.01).In group 4 the count of N in BALF was significantly lower than that in group 6(P<0.01).In group 2,3,4,5,6 and 7 the tidal volume(Vt),maximum voluntary ventilation(MVV),FEV0.3/FVC % and peak exhalation force(PEF)were significantly lower than those in group 1(P<0.01),and those in group 3,5,7 and 6 were significantly lower than those in group 2(P<0.01).In group 4,5 and 6 Vt was significantly higher than that in group 3(P<0.05).In group 4 MVV,FEV0.3/FVC% and PEF were significantly higher(P<0.05),and inspiratory resistance(IR)and expiratory resistance(ER)were significantly lower than those in group 3(P<0.05).In group 4 Vt and MVV were significantly higher(P<0.05),and IR and ER were significantly lower than those in group 7(P<0.05).Conclusion Tongsai Granules has inhibitory effect on the chemotaxis of WBC and N,and anti-infection effect.It can improve the pulmonary ventilation function in the model of COPD in acute exacerbation stage,and its effect is better than that of Tingbei Capsules.

摘要:Objective To investigate the interfering effect of curcumin(Cur)on smad signal transduction pathway of epithelial myofibroblast transdifferentiation(EMT)induced by transforming growth factor-β1(TGF-β1).Methods The human HK-2 cells were treated by TGF-β1(10 μg/L)at different time points(0,12,24,48 and 72 h).The expressions of α-smooth muscle actin(α-SMA)and E-cadherin were detected by using Western blotting method.The human HK-2 cells were pretreated with Cur in ascending concentration(1,5 and 10 μmoL/L)for 24 hours and then cultured in TGF-β1 culture solution for collecting and exacting protein and mRNA for 48 hours.The expressions of smad3,p-smad2/3,smad7 and TFβR-Ⅱ were detected by using Western blotting method,and mRNA expressions of collagen Ⅰ(Col Ⅰ)and Col Ⅲ were determined by applying RT-PCR.Results The phosphorylation of smad2/3 in HK-2 cells induced by TGF-β1 was earlier than the down-regulation of E-cadherin and the synthesis of α-SMA.After the intervention of Cur,the expressions of TFβR-Ⅱ,Col Ⅰ-mRNA and Col Ⅲ-mRNA,and phosphorylation of smad2 were inhibited significantly.The expression of smad7 was improved.Conclusion Cur can obstruct EMT process through interfering multiple sites of TGF-β1/smads signal transduction pathway.

摘要:Objective To observe the changes of glucocorticoid receptor(GR)and GR-mRNA in spleen cells before and after the intervene of glucocorticoid(GC)in mouse model of immune thrombocytopenic purpura and the regulation effect of Modified Xiaochaihu Granules(MXG).Methods All mice were divided into 5 groups,including the normal group,model group,MXG group,hormone group and MXG+hormone group.The mouse model of immune thrombocytopenic purpura was established in all groups except the normal group.After 7 days all groups were treated with the relevant drugs except the normal group(30 mL/kg,once a day for 14 days).After 21 days the mice were killed and the spleen was collected and nomogenized.The number of loci of spleen GR was deterimed with radioligand blinding assay,and the expression of GR-mRNA was detected with real-time fluorescence quantitative reverse transcription-polymerase chain reaction(RT-PCR).Results The number of loci of GR and GR-mRNA expression decreased in the model group with a significant difference compared with those in the normal group(P<0.05),increased in the MXG group with a significant difference compared with those in the model group(P<0.05),decreased more significantly in the hormone group with a significant difference compared with the model group(P<0.05),increased in the MXG+hormone group with a significant difference compared with the hormone group(P<0.05).Conclusion The number of loci of GR is lower,especially at the level of mRNA in mouse model of immune thrombocytopenic purpura.After the intervention by hormone the decrease of GR is more significant.Modified Xiaochaihu Granules can up-regulate the number of loci of GR,which performs at the level of mRNA.
摘要:Objective To compare the influences of Yiqihuoxue Formula(using Naoluoxintong,YF)and Bushenshengsui Formula(BF)on the expressions of Nestin in the subventricular zone(SVZ)of ischemic lateral ventricle,neuron specific enolase(NSE)and glial fibrillary acidic protein(GFAP)in the ischemic half-dark zone in rats with focal cerebral ischemia-reperfusion(I/R).Method The rat model of focal cerebral I/R was established by using thread approach,and then the rats were randomly divided into the sham-operation group,model group,YF group and BF group.After cerebral ischemia for 2 hours the reperfusion was given to the rats for 1 week,2 weeks or 3 weeks respectively.The positive cell quantity and the value of average absorbance(A value)in the expressions of Nestin,NSE and GFAP were detected by using immunofluorescence staining method.Result In SVZ the positive cell quantity in Nestin expression was higher significantly in YF group after I/R for 1 week and 2 weeks,and in BF group after I/R for 2 and 3 weeks than that in the model group(P<0.05 or P<0.01),that in YF group after I/R for 1 week was higher significantly than that in BF group(P<0.01),and that in BF group after I/R for 2 and 3 weeks was higher significantly than that in YF group(P<0.05).In the ischemic half-dark zone the positive cell quantity in NES expression was higher significantly in YF group after I/R for 1 week and in BF group after I/R for 2 weeks than that in the model group(P<0.01),that in YF group after I/R for 1 week was higher significantly than that in BF group(P<0.01),and that in BF group after I/R for 2 weeks than that in YF group(P<0.01).In the ischemic half-dark zone the average absorbance of GFAP expression was higher significantly in YF group after I/R for 1 week and 2 weeks and in BF group than that in the model group(P<0.05 or P<0.01)).There was no significant difference between YF group and BF group at all time points(P>0.05).Conclusion YF and BF can promote the proliferation and differentiation of neural stem cells after focal cerebral ischemia through increasing Nestin quantity in SVZ and NSE quantity,and improving GFAP expression in the ischemic half-dark zone.These two formulas have difference in the increase of Nestin and NSE quantity.

摘要:Objective To discuss the influence of resveratrol on the growth and cell cycle of human ovarian cancer cells(OVCAR3),and to study its inducing effect on the apoptosis of OVCAR3 and the possible pathway.Methods The influences of resveratrol on the growth,karyopycnosis and stegmentization were detected or observed by applying respectively MTT test,fluorescence microscope after Ho33342 staining,and the apoptosis of OVCAR3 was detected by using TUNEL.The flow cytometry was used to analyze the effects of resveratrol on cell cycle distribution and activity of caspase-3.Results The proliferation of OVCAR3 was promoted by resveratrol in low concentration(<30 μmol/L)and inhibited in high concentration(>30 μmol/L).The retardation of S/G2 cells and OVCAR3 apoptosis could be induced by resveratrol.The increase of caspase-3 activity induced by resveratrol showed a time-efficiency and concentration-efficiency.Conclusion Resveratrol can improve or inhibit the growth of OVCAR3 in a manner of concentration dependency,and it can induce OVCAR3 apoptosis through promoting caspase-3 activity.
摘要:Objective To observe the changes of hydrogen sulfide(H2S)and cystathionine-γ-lyase(CSE)in the course of resisting acute lung lesion(ALL)induced by lipopolysaccharide(LPS)with Dachnegqi Decoction in aged rats.Methods SD adult rats(n=88)were randomly divided into the sham-operation group,ALL model group(established by intratracheal instillation of LPS to induce ALL),propargylglycin(PPG,an inhibitor of CSE)group and Dachengqi Decoction group.After medicine administration for 4 and 8 hours the rats were killed and the lung coefficients were determined respectively.The morphological changes in the lung tissue were observed with optical microscope,the plasma content of H2S and malondialdehyde(MDA),and the activities of myeloperoxidase(MPO)and CSE were detected by using chemical method.The changes of the expression of CSE-mRNA were detected by applying RT-PCR.The correlation between plasma H2S and the above indexes were analyzed.Results The obvious morphological changes in the lung tissue were induced by the intratracheal instillation of LPS.The lung coefficients,content of MDA and activity of MPO were improved.The plasma content of H2S,CSE activity and CSE-mRNA expression decreased.The pre-administration of PPG aggravated the lung lesion induced by LPS,and Dachengqi Decoction relieved the lung lesion,reduced lung coefficients,MDA content,and MPO activity,and at the same time increased the plasma content of H2S,CSE activity and CSE-mRNA expression.The outcomes of correlation analysis showed that H2S was correlated to CSE activity(r=0.950-0.981,all P<0.01),and negatively related to other indexes(r=-0.985—-0.933,all P<0.01).Conclusion The down-regulation of H2S-CSE system takes part in the onset of ALL induced by LPS in aged rats.The up-regulation of H2S-CSE system is one of mechanisms of Dachengqi Decoction resisting ALL induced by LPS in aged rats.
摘要:Objective To study the antidepressant effect of Zhizhukuanzhong Capsules on animal model of depression.Methods The tests of forced swimming,tail suspension,antagonism of reserpine and accentuation of 5-HTP in mice and chronic stress in rats were carried out to observe the antidepressant effect of Zhizhukuanzhong Capsules.Results Zhizhukuanzhong Capsules(1.106 g/kg)shortened the dead time of tail suspension and forced swimming in mice.Zhizhukuanzhong Capsules in different doses resisted the descent of body temperature induced by the injection of reserpine after one hour,while in dose of 1.106 g/kg increased the frequency of head flick in mice.In chronic stress rats the horizontal movement was improved by Zhizhukuanzhong Capsules in dose of 0.774 g/kg,glycotropic degree was increased in dose of 0.774 g/kg or 0.387 g/kg,serum content of corticosterone was reduced,and the expression of 5HT1A receptor was promoted.Conclusion Zhizhukuanzhong Capsules has antidepressant effect and can relieve the depressant behaviors of animals.The mechanism may be related to that it improves 5HT neuron transmission and inhibits the secretion of adrenocortical hormones.

摘要:Objective To investigate the influence of inflammatory reaction on the left ventricular remodeling induced by spontaneously hypertension,and to discuss the intervene of Jiangyamaijing Solution.Methods The rats with spontaneously hypertension(8 weeks old,n=40)were randomly divided into the normal control group(group A),model group(group B),treatment group treated with Jiangyamaijing Solution and Luohuoxi Tablets orally(group C),Western medicine control group treated with Luohuoxi Tablets orally(group D)and Chinese medicine control group treated with Niuhuangjiangya Pills and Luohuoxi Tablets orally(group E).One treatment course was 20 weeks.The blood pressure and pathological changes were observed,the heart weight index(HWI),myocardial interstitial collagen volumetric fraction(CVF)and perivascular collagen area(PVCA)were calculated,and the expressions of serum interleuking-6(IL-6)and tumor necrosis factor-α(TNF-α)were detected by using radioimmunoassay.Results ①In group D the cardiomyocyte hypertrophy and myocardial interstitial inflammatory cell infiltration were relieved and collagen content was reduced significantly(P<0.01).HWI had a trend of descent.②The expressions of serum IL-6 and TNF-α were higher significantly(P<0.01)in rats with spontaneously hypertension than those in normal rats.③In group D the expression of serum IL-6 decreased significantly compared with that in group B and the effect was better than that in other groups(P<0.01).Conclusions The excess expressions of serum IL-6 and TNF-α may be involved in the left ventricular remodeling induced by spontaneously hypertension.The remodeling can be reversed by Jiangyamaijing Solution and the mechanism may be related to the decrease of IL-6 expression.

摘要:Objective To investigate the influences of common preparation methods of slices of Rhizoma Chuanxiong on the changes of their chemical compositions by applying fingerprint chromatogram,and their dissolution characteristics in the decoction of the slices of Rhizoma Chuanxiong after exacting for different times.Methods The changes of chemical compositions of the slices,and difference between extractable compositions and original compositions of Rhizoma Chuanxiong were compared before and after the preparation by applying HPLC fingerprint chromatogram and the control and analytic method of chemical standard samples.Results The chemical compositions of the slices was not much different from original compositions after preparation.The liposoluble compositions of the slices was dissolved less in the decoction,while the most of water-soluble compositions were extracted out after extracting for three times.Conclusion When the slices of Rhizoma Chuanxiong were decocted alone,less liposoluble compositions are dissolved in decoction,which indicates that the effective material base(chemical compositions)of the slices of Rhizoma Chuanxiong in decoction should be water-soluble compositions.

摘要:Objective To set up a method for determining and analyzing baicalin,chlorogenic acid and jasminoidin in Kangganjiedu Oral Solution simultaneously by multi-wavelength RP-HPLC.Methods The procedure of multi-wavelength RP-HPLC was performed on the chromatographic column of Agilent Zorbax SB C18(250 mm×4.6 mm,5 μm).The mobile phase of linear gradient elution was 0.3% aqueous formic acid solution(A)to acetonitrile(B),and performed as follows:B:14%-20%,0-12 min;B:20%-30%,12-13 min;B:30%-65%,13-25 min;B:65%,25-35 min.The flow rate was 1.0 mL/min.The column temperature was 30 ℃.Results Baicalin showed a good linear relationship at a range of 0.225-3.60 μg,chlorogenic acid,at 0.023 0-0.368 0 μg,and jasminoidin,at 0.135-2.16 μg.The average recovery of baicalin,chlorogenic acid and jasminoidin was 99.62%,99.64% and 100.3% respectively.RSD was 0.68%,1.5% and 0.23% respectively.Conclusion The method was easy and accurate with a higher repeatability,which can be used in the determination of baicalin,chlorogenic acid and jasminoidin in Kangganjiedu Oral Solution simultaneously.
摘要:Objective To study the technology condition of extracting Scolopendra by bionic enzymatic method.Methods The technology condition of extracting Scolopendra by bionic enzymatic method was observed by single factor observation taken the hydrolysis degree as the index,and the extractive technique was verified by using the methods of active partial thromboplastin time(APTT)and fibrinogen plate.Results The coagulation time was longer and anti-thrombosis effect was more significant in the bionic enzymatic method.The best enzymatic condition was as follows:Scolopendra was hydrolyzed in artificial gastric juice(37 ℃)and pepsin(ratio of pepsin to substrate was 4.0%)for 4 hours,and then the residue was hydrolyzed in artificial intestinal juice(37 ℃)and trypsin(ratio of pepsin to substrate was 5%)for 4 hours.Conclusion The bionic enzymatic method can be used in the extraction of effective components of anti-coagulation and thrombolysis from Scolopendra.
摘要:Objective To investigate the characteristics of syndrome distribution and syndrome factor combination of advanced non-small cell lung cancer(NSCLC)of preliminary diagnosis.Methods A review survey was conducted to collect 70 items of information by TCM four diagnostic methods from 283 cases of advanced NSCLC hospitalized in our hospital from Jan.2002 to Nov.2007.Every case was classified according to the syndrome differentiation and given descriptive statistical analysis.Results There were 25 types of syndromes in 283 cases of advanced NSCLC.The occurrence rates of types of qi-yin-deficiency,qi-deficiency,qi-deficiency and blood-stasis,and phlegm-dampness obstruction were higher.The main syndrome factors included qi-deficiency and blood-stasis counted for 65.7% and 48.4% respectively,and other factors are yin-deficiency,phlegm-dampness,fluid-retention in the chest and hypochondrium,phlegm-heat,qi-stagnation and yang-deficiency in a descending order.The common one-one combinations of syndrome factors included qi-deficiency and blood-stasis,qi-deficiency and yin-deficiency,phlegm-damp and blood-stasis,qi-deficiency and phlegm-damp,and yin-deficiency and blood-stasis in a descending order.Conclusion The syndrome distribution of preliminary diagnosed NSCLC was complicated and multiple.The train of thought of syndrome factor combination accords with the clinical practice of advance NSCTC from the aspect of the factors of syndrome differentiation.
摘要:Objective To observe the curative effect of syndrome differentiation-based treatment with needle-scalpel therapy on knee osteoarthritis.Methods The patients were randomly divided into the treatment group(locating acupoints by syndrome differentiation)and control group(locating acupoints by local pain)and all treated with the needle-scalpel therapy.The scores of curative effect in two groups were calculated by using Lequesne serious index of knee osteoarthritis before and after the treatment.Results In two groups the scores all decreased significantly before and after the treatment respectively(P<0.01),while the score and superior rate in the treatment group were higher significantly than those in the control group(P<0.05).Conclusion The needle-scalpel therapy has a better short-term curative effect on knee osteoarthritis after syndrome differentiation.
