摘要:The characteristics of syndromes are manifested as periodic changes of lung,spleen and kidney found in the development process of exopathic diseases through analysing on syndrome differentiation and treatment laws of exterior syndrome in dual disease of exterior and interior in Shanghanzabinglun. Combining with the theory of"defense qi originating from triple energizer",the author puts forward the view of"exterior syndrome treated based on triple energizer",i.e.the lung is the first defensive line, which should be assisted to diffuse defense qi by using the kind of Chinese herbal remedies(CHR) being pungent in flavor with the action of dispersing such as Mahuang(Herba Ephedrae);the spleen is the second defensive line,which should be assisted to nourish the source of defense qi by using the kind of CHR being sweet in flavor and warm in nature with the action of tonifying and replenishing the middle qi such as Guizhi(Ramulus Cinnamomi);and the kidney is the third defensive line,which should be assisted to secure the root of defense qi by using the kind of CHR being pungent in flavor and hot in nature with the action of assisting yang such as Fuzi(Radix Aconiti Lateralis Praeparata).The view of"exterior syndrome treated based on triple energizer"not only guides clinical practice more exactly,but also meets the holism concept of Chinese medicine.

摘要:Objective To establish the vascular endothelial cell lesion model of blood-stasis syndrome of type 2 diabetes mellitus.Methods ECV-304 ceils of logarithmic phase were intervened and divided into the blank control group(group 1),health group(group 2),blood-stasis syndrome of diabetes mellitus group(group 3) and diabetes mellitus group(group 4).The activities of ECV-340 cells were observed by using MTT test.The morphological changes of the cells were observed by inverted phase contrast microscope,scanning electron microscope and transmission electron microscope.The levels of ET and NO were detected by using radioimmunoassay and nitrate reductase method respectively.The levels of endothelial protein C receptor(EPCR),yon Willebrand factor(vWF) and thrombonodulin(sTM) were detected by applying double-antibodies-sandwich-ELISA method.The changes of free calcium in the cells were observed by laser scanning confocal microscope(LSCM) taken Fluo-3/AM as fluorescence indicator.The distribution of actin microfilament was observed by using rhodamine phalloidin staining. Results①The value of optical density(OD) decreased significantly in group 3 than that in group 2 (P<0.001).②In group 3 ET level was higher than that in group 1 and that in group 4(P<0.001), NO level was lower than that in group 1(P<0.001) and had no statistical difference compared with that in group 4(P>0.05).③In group 3 EPCR level was higher than that in group 1 and lower than that in group 4(P<0.001),the level of vWF was higher than that in group 1(P<0.001) and had no statistical difference compared with that in group 4(P>0.05),and sTM level was higher than that in group 1(P<0.001) and had no statistical difference compared with that in group 4(P>0.05).④In group 3 the distribution of fluorescence showed uneven and fluorescence intensity was higher than that in group 1 and group 4(P<0.001).⑤In group 1 and 2 the distribution of microfilament cytoskeleton was regular and clear with normal cell shape,while in group 3 it was broken and irregular,and in group 4 it was broken but regular.Conclusion ECV-304 cells showed various signs of vascular endothelial cell lesion after the interference by the serum from the patient with type 2 diabetes mellitus.The vascular endothelial cell lesion model of blood-stasis syndrome combined disease and syndrome was established initially.

摘要:Objective To analyse the effect of Naoshuning on brain edema through observing the dynamic changes of magnetic resonance imaging(MRI) in rats with brain contusion and laceration.Methods All 114 male SD rats were randomly divided into Naoshuning group,model group and sham-operation group. The rat model of brain contusion and laceration was established by free-falling impact according to Feeney’s method.After the operation the dynamic changes of MRI signal intensity(SI) and volume in T2 WI focus were observed consecutively,and the brain water content was determined by using dry-wet method.Results There were changes observed in increased SI of T2 in the cerebral hemisphere with operation.SI got the highest level and volume on the first day and then decreased gradually and reached the lowest level on the seventh day,which showed a parallel course with brain water content.In Naoshuning group SI in T2 WI focus,edema volume and brain water content were significantly lower than those in the model group at all time points(P<0.05).The absorption rate of edema in Naoshuning group was significantly higher than that in the model group(P<0.005) seven days after the operation. Conclusion The non-invasive MRI examination can be used to analyse quantitatively the degree of brain trauma and edema in rats after the treatment with Naoshuning,which is reliable for review the curative effect.Naoshuning can improve the absorption of edema after brain contusion and laceration.
摘要:Objective To study the influence of Siwu Mixture on metabolites in rats after the intraperitoneal injection of chemotherapeutics-Cisplatin.Method The rats(n=6) were randomly divided into the experimental group,model group and normal group(each n=2).The experimental group and model group were respectively given Siwu Mixture and purified water intragastrically after the intraperitoneal injection of Cisplatin,and the normal group was given purified water intragastrically after the injection of normal saline solution.The urine were collected from the rats in all groups every day and the spectroscopy of 1H-nuclear magnetic resonance(1H-NMR) was determined.Result After multivariate statistical analysis the metabolites of the experimental group,model group and normal group were different significantly,especially acetic acid,alanine,lactic acid and hippuric acid.Conclusion The result of the study indicates that Siwu Mixture can protect the functions of the liver and kidney.

摘要:Objective To observe the influences of Ganfujian Formula on the expressions of transforming growth factor-β1(TGF-β1),TβRII-mRNA,Smad2/3,Smad4 and Smad7 in rats with hepatic fibrosis induced by porcine serum.Methods The hepatic fibrosis model was established in SD rats by the intraperitoneal injection of porcine serum(0.5 mL,twice a week for 8 weeks).The rats then were given Ganfujian Formula orally in different doses(1.50 g/kg,2.25 g/kg and 3.0 g/kg) and colchicines (0.154 g/kg) respectively until the end of 12th week.The expressions of TGF-β1 and TβRII-mRNA were detected by using RT-PCR and the expressions of Smad2/3,Smad4 and Smad7 were detected by applying Western blotting method.Results In the model group,compared with the normal control group,the expressions of TGF-β1 and TβRII-mRNA,Smad2/3 and Smad4 increased significantly and Smad7 expression decreased significantly.After the treatment with Ganfujian Formula the expressions of TGF-β1 and TβRII-mRNA were reduced in the treatment group compared with the model group. Ganfujian Formula regulated downward the expressions of Smad2/3 and Smad4,and regulated upward Smad7 expression at the same time.Conclusion Ganfujian Formula can inhibit the expressions of TGF-β1 and TβRII-mRNA,regulates downward the expressions of Smad2/3 and Smad4,and regulates upward Smad7 expression.It may be its one of the mechanisms for curing hepatic fibrosis that it has an obvious influence on hepatic TGF-βand Smad signal pathway.

摘要:Objective To observe the influences of astragalus polysaccharide(APS) on hydroxyproline and matrix metalloproteinase-9(MMP-9) in rats with chronic obstructive pulmonary disease(COPD). Methods The rat model of COPD was established by using the method of cigarette smoking combined with intratracheal injection of lipopolysaccharide(LPS).APS(in dose of high,middle and low) and prednisone were given to the rats intragastrically for intervention for 30 days.The lung content of hydroxyproline was detected by using biochemistry method,the gene expression of MMP-9 was detected by applying reverse transcription polymerase chain reaction(RT-PCR),and the protein content of MMP- 9 was determined semi-quantitatively by using immunohistochemistry method.Results In the model group the content of hydroxyproline and MMP-9 expression increased significantly compared with those in the normal control group(P<0.01).After the intervention of APS the content of hydroxyproline decreased significantly in a manner of dose-dependent,and the gene expression and protein level of MMP-9 decreased significantly,especially when APS was in middle dose.Conclusion APS can decrease the lung content of hydroxyproline and MMP-9 expression in rats with COPD,which may be related to that APS can relieve the lung lesion.

摘要:Objective To study the effect of rhynchophylline(Rhy) on carotid sinus baroreflex(CSB) and the mechanism.Methods The technique of carotid sinus isolation and perfusion was used to determine the parameters of CSB,including threshold pressure(TP),saturation pressure(SP), equilibrium pressure(EP),operating range(OR),peak slope(PS) and reflex decrease(RD).The influence of Rhy on CSB was observed in a slope manner after Rhy(10,50 and 100μmol/L) dissolving in K-H solution and then filling into carotid sinus.Nitric oxide synthase inhibitor[NG-nitro-arginine methyl ester(L-NAME)],potassium channel blocker[tetraethylammonium(TEA)]and agonist of L-type calcium channel(Bay K8644) were perfused into isolated carotid sinus before reperfusion of Rhy, and then the effects of them on Rhy were observed.Results Rhy inhibited CSB in a manner of dosedependent with the increase of TP and SP and the decrease of PS and RD.The inhibitory effect of Rhy was not affected by the pretreatment of L-NAME(300μmol/L) or TEA(1 mmol/L),but attenuated by Bay K8644(500 nmol/L).Conclusion Rhy inhibits CSB,which may be related to that Rhy attenuates calcium influx of mechanosensitive ion channel of baroreceptor but not related to NO channel or potassium channel.

摘要:Objective To study a method for the quality control of extracts from Zhizi(Fructus Gardeniae).Methods The quality control of extracts from Zhizi was carried out through thin-layer chromatography(TLC),examinations of moisture content,residue of ignition,heavy metals and arsenic salt,and ultraviolet spectrophotometry and HPLC.The main chromatographic peaks of fingerprint were identified.Results The experimental sample and control sample in TLC showed the same colored spots at the same position(negative without interference).The results of all examinations were accorded with the China National Pharmacopoeia.In the extracts from Zhizi the total iridoid glycosides showed a good linear relationship at a range of 5.592 - 35.712μg(r=0.999 9),the recoveries of the samples in the concentrations of high,middle and low were 98.34%,100.67%and 101.07%respectively,and RSD were 2.60%,1.47%and 1.20%respectively(n=3).Geniposide showed a good linear relationship at a range of 0.228 - 1.140μg(r=0.999 9),the recoveries of the samples in the concentrations of high, middle and low were 104.04%,100.97%and 95.94%respectively,and RSD were 0.17%,0.58%and 0.84%respectively(n=3).Conclusion TLC and HPLC-MS can be used to identify qualitatively the chemical constituents of extracts from Zhizi,ultraviolet spectrophotometry can be used to control the quantity of the total iridoid glycosides,and HPLC can be used to control the quantity of geniposide.The method is simple,accurate with a higher repeatability.

摘要:Objective To investigate the chemical constituents of Zicao(Radix Arnebiae) produced from Changbai Mountain.Methods Multi-chromatographic methods including reflux of petroleum ether,silica gel column chromatograph,preparative TLC and recrystallization were used to isolate the extracts from Zicao to get the monomeric compounds.The chemical structures of the extracts were identified based on the physicochemical characters and spectroscopic data.Results There were 5 chemical compounds extracted from Zicao,including deoxyshikonin,β,β’-dimethyl-acryl-shikonin,acetylshikonin,teracryl-shikonin and shikonin.Conclusion Deoxyshikonin,β,β’-dimethyl-acryl-shikonin,acetylshikonin, teracryl-shikonin and shikonin are found in Zicao produced from Changbai Mountain for the first time. The chemical constituents identified in Zicao produced from Changbai Mountain are the same as those in authentic Zicao.
摘要:Objective To select the ultrafiltration membrane for removing pyrogen from Qingkailing Injection.Methods The tachypleus amebodyte lysate(TAL)-gel method was used to determine a pore size of ultrafiltration membrane for removing pyrogen from Qingkailing Injection after initial screening of the capability of the membrane in elimination of bacterial endotoxin.The pyrogen was detected by pyrogen test to verify the effect of pyrogen elimination of the membrane when Qingkailing Injection passed through.The processing condition of the membrane was screened based on the transfer rate and membrane flux of baicalin(index constituent).Results TAL with the sensitivity of 0.06 EU/mL was used in the membrane examination.The drink water was changed to meet the standard of injection water after passing through the membrane of 10 KD and 30 KD.In the pyrogen examination of 6 batches of Qingkailing Injection before ultrafiltration the minimum sum of elevated temperature was 1.04℃and maximum was 2.40℃in three rabbits.After ultrafiltration by the membrane of 30 KD the minimum sum of elevated temperature was 0.43℃and maximum was 0.88℃, which was significantly lower than 1.0℃.When the temperature of Qingkailing Injection was 40℃,pump speed was 250 r/min,and working pressure was 0.10-0.15 MPa,the ultrafiltraion flux was the highest with maximum efficient.Conclusion The ultrafiltration membrane of 30 KD can be used to eliminate pyrogen from Qingkailing Injection effectively.The temperature and pump speed have influences on membrane flux significantly.The transfer rate of baicalin is 97.15%after ultrafiltration,which indicates that the content of index constituent meets the quality standard of Qingkailing Injection.

摘要:Objective To investigate the correlation between the coagulation factors and TCM syndrome elements of acute cerebral infarction(ACI).Methods The syndromes of 148 patients with ACI were scored according to Diagnostic Criteria for Syndrome Differentiation of Stroke(Trial Implementation).If the score was higher or equal to 7,the diagnosis was tenable.There were 6 syndrome element groups including wind,fire,phlegm,blood stasis,qi deficiency and yin deficiency with yang hyperactivity.The prothrombin time(PT),activated partial thromboplastin time(APTT) and fibrinogen(FIB) were detected and the correlation between TCM syndrome elements and coagulation factors was studied by applying rank-correlation analytic method.Results The value of PT in qi-deficiency group was lower than that in blood-stasis or wind group(P<0.05),and was not different compared with that in other groups.The value of APTT or FIB was not different among all groups(P>0.05).There was a negative correlation between values of FIB and wind or fire syndrome element(P<0.05),and a negative correlation between values of PT and phlegm or qi-deficiency syndrome element(P<0.05,P<0.01). Conclusion The higher of the scores of wind and fire elements were,the lower of FIB was,and the patient may be in hypocoagulable state.The higher of the scores of phlegm and qi-deficiency elements were,the lower of PT was,and the patient may be in hypercoagulable state.
摘要:Objective To observe the curative effects of the self-made decoction with the actions of warming kidney and draining dampness,and activating blood circulation and descending turbid on chronic renal failure(CRF).Methods The patients(n=356) with CRF were divided,according to the principle of parallel control,into the treatment group(n=187) treated with the decoction(one decoction daily) combined with Western expectant treatment and control group(n=169) treated with Western expectant treatment only.After 6 months the grade quantity integrals of symptoms were observed and some indexes including kidney function,platelet and coagulation function were detected.Results In the treatment group the kidney function and symptom integrals were improved significantly and had statistical significance compared with those of the control group(P<0.05 or P<0.01).Conclusion The self-made decoction with the actions of warming kidney and draining dampness,and activating blood circulation and descending turbid can relieve the symptoms of the patients,improve the kidney function and delay the development of CRF.

摘要:Objective To investigate the curative effect of Ruanmailing Oral Solution on vascular dementia and its influences on the levels of superoxide dismutase(SOD),vascular endothelial growth factor (VEGF),estradiol(E2) and testosterone(T) in the patients with vascular dementia(VaD).Methods All patents(n=60) were randomly divided into Ruanmailing Oral Solution group(treatment group) and Sibohai Tablets group(donepezil hydrochloside,control group).The treatment group was given Ruanmailing Oral Solution thrice daily and one piece(10 mL) each time for 3 treatment courses(a course was for one month) by applying the single blind method.The control group was given Sibohai Tablets once daily and one table(5 mg) each time at sleeping time with the same course as the treatment group.The normal control group included 30 health old persons.The levels of SOD,VEGF,E2 and T were detected, and the integral changes of the Mini-Mental Status Examination(MMSE),Activities of Daily Life(ADL) and TCM symptoms were observed.Results Ruanmailing Oral Solution increased significantly the levels of SOD and VEGF(P<0.05),E2 value in woman patients and T value in man patients,and decreased E2 value and E2/T value in man patients(P<0.05 or P<0.01).The integral of MMSE was increased and ADL integral was decreased.The clinical symptoms were relieved.Conclusion Ruanmailing Oral Solution can be used to prevent and cure VaD effectively.
摘要:Objective To study the characteristics of tongue fur in the cases of HIV/AIDS by applying clinical epidemiological method in order to discuss the cause and pathogenesis.Method The random sampling method of multiple-stage stratified cluster was used to collect and analyse the characteristics of tongue fur in 1 323 cases of HIV/AIDS and 778 control cases.Result Among the characteristics of tongue fur in the cases of HIV/AIDS yellow tongue fur accounted for 36.4%,yellow and white fur, 12.8%,slimy fur,45.4%,thick fur,40.2%,rotten-curdy fur,0.9%,and powder-like fur,1.3%. The incidence rate of the above abnormal fur at mid-root tongue was higher in the cases of HIV/AIDS than that in the control cases(P<0.05).Conclusion The result of the study indicates that the pathogen of AIDS has the characteristics of dampness-turbid and heat-toxin.
