      摘要:The course of TCM diagnosis and treatment is a dynamic,continues and complicated cognitive one,and it is also a repeated conformation by physician on patient’s disease.Different physicians may give different diagnoses or different conformations on the same disease according to their knowledge background and clinical experiences.The difference of cognition influences curative effects in clinic.A physician plays a key role in the course of TCM diagnosis and treatment.The paper analysed the formation of physician’s thinking conformation,and suggested to establish a systemic model on computer by a integrative method of quality and quantity,which provides a practicable platform for transforming physician’s latent knowledge into dominant one,and expand and definite his thinking of diagnosis and treatment at a greater width.

      摘要:The paper provided a quantitative method of Chinese herb nature using 24 medicinal natures as the basic index of single herb.The concept of formula nature was proposed and a mathematic presentation method was made out.The formulas with cool or warm nature and the formula used to treat wasting-thirst were studied based on the concept of formula nature.The results indicated that the concept of formula nature can reflect the nature composition of different kind of formulas.The study gives a new idea in the further research of formula design laws.

      摘要:The secondary development of high quality well-know formulas is a new way to improve the active development of TCM,which is important to the modernization of TCM and growth of Chinese medical cause.The paper discussed the thought and way of the secondary development of high quality well-know formulas taken refined Qingkailing Injection as an example based on new medicinal research,TCM theory,selection of formulas and diseases,formula optimization and mechanism research.The author believed that the secondary development should follow the above guideline,and also pay attention to the guiding significance of TCM theory.

      摘要:Objective To investigate the distribution characteristics of common TCM syndromes and syndrome factors of ulcerative colitis(UC).Methods The literature were selected from 1979 to 2007 through retrieval,and then a database was established and analysed statistically.Results There were totally 443 syndrome types from the literature and 93 standardized.The syndromes with the frequency higher than 5% included,in order,spleen-kidney yang deficiency,dampness heat,liver depression and spleen deficiency,spleen-stomach weakness and blood stasis due to qi stagnation.There were 28 syndrome factors obtained,and among them 11 pertained to location factors including spleen,kidney,liver,stomach and intestines with the frequency higher than 1%,and 17 pertained to nature factors including qi deficiency,dampness,heat,yang deficiency,qi stagnation,blood stasis,cold,yin deficiency and blood deficiency with the frequency higher than 1%.Conclusion The syndrome types of UC are complicated in clinic and syndrome factors are relatively simple.The study on common syndromes of a disease based on syndrome factors will go from the simple to the comlex.

      摘要:Quanyingfanglun,written by ZHENG Duan-you of the Southern Song Dynasty,is a medical book with clinical achievements.The collection of hand-written Quanyingfanglun is kept in Japan and is not kept in China.The rare book stack-room of the library of China Academy of Chinese Medical Sciences,however,collects the fragments of Baoyingquanfang.It is conformed that the two books are synonym after repeated textual research.There are illustrations in the beginning of volume one of Quanyingfanglun drawing five zang-viscera invading by diseases,which are clearer than those in the contemporary book-Youyouxinshu.These illustrations are accompanied by poetries elaborating the methods of syndrome differentiation through observing complexion and finger venules.The similar illustrations were reprinted in many child medical books of the Ming and Qing Dynasties,which indicates that the book had been valued by physicians of the Song Dynasty and has been inherited by later physicians.In last two volumes of the book there are 31 medical records recorded by ZHENG Duan-you,among them there are 8 about risk diseases with poor prognoses.It is helpful for diagnosis and treatment of child diseases.

      摘要:Objective To discuss the mechanism of Xiaoyao Powder in the treatment of liver depression and spleen deficiency syndrome induced by chronic stress.Methods All 100 male SD rats were randomly divided into the normal group,sham-operation group,model group,Xiaoyao Powder group,CNQX group and Xiaoyao Powder combining with CNQX group(combining group).The model of liver depression and spleen deficiency syndrome was established in rats by chronic bondage stress for 21 days.The Xiaoyao Powder group and combining group were given Xiaoyao Powder(5.32 g/kg) orally every day before bondage.The CNQX group and combining group were injected with CNQX(a receptor antagonist of AMPA,0.5 μg) in the right amygdale on the 1st,4th,7th,10th,13th,16th,19th and 21st day,respectively.The open-field test and elevated plus-maze test were carried out in rats of all groups on the 22nd day.Results In the open-field test the stay period in the central zone increased significantly,and the total distance of 5-minute motion decreased significantly in the model group.In the elevated plus-maze test the times and stay period in the central zone,and times and stay period of entering open arm decreased significantly,and the stay period in close arm increased significantly in the model group.All indexes were improved significantly by CNQX or Xiaoyao Powder in the open-field test and elevated plus-maze test.Xiaoyao Powder combining with CNQX had better effect.Conclusion Xiaoyao Powder can relieve the ethologic changes of rat model of liver depression and spleen deficiency syndrome,and the mechanism is as the same as that of CNQX.

      摘要:Objective To investigate the mechanism of aralosides in antagonism of myocardial ischemia,and provide a theoretical base for it used for preventing and treating coronary heart disease(CHD).Methods All rats were randomly divided into the control group(0.9% sodium chloride solution,10 mL/kg),aralosides group(aralosides solution,0.5 g/kg) and clopidogrel group(clopidogrel solution,5 mg/kg).After intragastrical administration of drugs for 5 days respectively in all groups,the anterior descending coronary was ligated to induce ischemia for 45 minutes and then reperfused for 5 minutes.The pathological changes of myocardial tissue were observed in all groups,the expression of endocardial CD40L was detected by using immunohistochemistry,and levels of myocardial malondialdehyde(MDA) and glutathione peroxidase(GSH-PX) were detected.Results Compared with the control group,the expression of CD40L decreased(P<0.05),and vigour of myocardial GSH-PX increased(P<0.05) in aralosides group and clopidogrel group,but the difference was not significant statistically(P>0.05).The level of myocardial MDA decreased in the aralosides group(P<0.05).Conclusion Aralosides can decrease the expression of CD40L,improve the vigour of myocardial GSH-PX and reduce the level of myocardial MDA in rats with myocardial ischemia and reperfusion.

      摘要:Objective To study the pathological changes of cerebral capillary in KKAy mice,and determine the specific manifestations of brain collateral disease of diabetes.Methods Male KKAy mice(9-11 weeks old) were included into the model group and male C57BL/J mice(9-11 weeks old) were included into the control group.The fasting plasma glucose and body weight were measured in mice of 16,20,24 and 28 weeks old respectively,and on the 28th week all mice were sacrificed.The serum content of 6-Keto-PGF1α and TXB2 were detected.The brain pathomorphological changes were observed with the light microscope and transmission electron microscope(TEM).Results Compared with C57BL/J mice,in KKAy mice the levels of weight and plasma glucose were higher,serum content of TXB2 increased and serum content of 6-Keto-PGF1α deceased.The nuclei of brain capillary endothelial cells were loose and swollen,lumen became narrow with stasis of erythrocytes and platelets,nuclei and mitochondria of nerve cells were swollen,rough endoplasmic reticulum became extenuation,and ribosome reduced.Conclusion The structure and function of brain capillary endothelial cells are injured by higher plasma glucose and abnormal serum content of 6-Keto-PGF1α and TXB2,which will induce the destroy of neurocyte structure finally.

      摘要:Objective To study the influence of Kudiezi Injection,having the actions of detoxification and dredging collaterals,on the expression of Rho-associated coiled-coil containing protein kinase α(ROKα) in rats with acute cerebral ischemia,and to investigate the brain protection mechanism of the therapy of detoxification and dredging collaterals.Methods The expression and distribution of brain ROKα were detected by using immunohistochemistry in rats with focal cerebral ischemia induced by middle cerebral artery occlusion(MCAO).The expression of ROKα-mRNA and its changes after intervened by Kudiezi Injection or Hydroxyfasudil Injection(a selective inhibitory agent of ROK) were detected by applying real-time fluorescence quantitative polymerase chain reaction(RT-PCR).Results The expression of ROKα was high abnormally in ischemia brain of rat model(P<0.01).Kudiezi Injection decreased significantly the expressions of ROKα and ROKα-mRNA in ischemia cortex and hippocampus(P<0.01).The comparison between Kudiezi Injection and Hydroxyfasudil Injection did not show significant difference(P>0.05).Conclusion Kudiezi Injection,having the actions of detoxification and dredging collaterals,has a brain protective effect in rats with acute cerebral ischemia,and the mechanism may be related to that it decreases the expression of ROKα in ischemia brain.

      摘要:Objective To study the influences of Chinese herbal remedies,having the actions of replenishing qi,nourishing yin and activating blood circulation,and detoxification and activating blood circulation on early ventricular remodeling in rats after myocardial infarction and mechanism of the effects.Methods The model of acute myocardial infarction(AMI) was established by ligating the left anterior descending coronary in male Wistar rats.The rats were randomly divided into the sham-operation(with puncturing only without ligating,group B),model group(group C),perindopril group(0.36 mg/kg,group D),group of replenishing qi,nourishing yin and activating blood circulation(Shengmai Capsules 0.24 g/kg and Compound Chuanxiong Capsules 0.40 g/kg,group E) and group of detoxify and activating blood circulation(Coptis Alkaloid 0.16 g/kg and Compound Chuanxiong Capsules 0.40 g/kg,group F,each n=10),and given orally relevant medicinals respectively.Other 10 rats were included into the normal group(group A) and given equivalent water.After 4 weeks all rats were anesthetized and killed and the left ventricular mass index(LVMI) was detected.The serum levels of hypersensitive C-reaction protein(hs-CRP),interleukin-1β(IL-1β) and interleukin-6(IL-6) were determined.The expressions of nuclear factor-κB-mRNA(NF-κB-mRNA) and peroxisome proliferators-activated receptor-γ-mRNA(PPAR-γ-mRNA) in ischemia myocardium were detected by using RT-PCR.Results Compared with group C,the left ventricular weight and LVMI decreased in group D,E and F respectively(P<0.01;P<0.05).The serum level of IL-6 decreased significantly in group F(P<0.05).The expressions of PPAR-γ-mRNA and NF-κB-mRNA decreased significantly in group E and F(P<0.05),and the expression of PPAR-γ-mRNA was lower significantly in group F than that in group E(P<0.05).Conclusion The components of Chinese herbal remedies with the action of replenishing qi,nourishing yin and activating blood circulation,and detoxification and activating blood circulation can decrease the expressions of inflammatory genes by different ways in order to inhibit ventricular remodeling after AMI in rats.

      摘要:Objective To study the protein expression of myocardial cardiotrophin-1(CT-1) and mechanism of Huangqi Injection for relieving myocardial pathological changes in diabetic rats.Methods All male Wistar rats were randomly divided into the normal group(n=8) and diabetic model group(n=22),and the later was randomly divided again into the model group and Huangqi group(each n=11) after modeling.The Huangqi group was given intragastrically Huangqi Injection(8 g/kg·d) for 8 weeks.The pathological changes of heart index(heart weight/body weight) and myocardial fiber content were determined and the expression of CT-1 was detected by using immunohistochemistry.Results The level of plasma glucose was higher in the model group and Huangqi group than that in the normal group(P<0.01).The heart index was higher in the model group than that in the normal group(P<0.01),and was lower in the Huangqi group than that in the model group(P<0.05).The content of collagen calculated by target area in Masson staining was higher in the model group than that in the normal group(P<0.01),and lower in the Huangqi group than that in the model group(P<0.05).The expression of CT-1 was higher in the model group than that in the normal group(P<0.01),and lower in the Huangqi group than that in the model group(P<0.05).Conclusion The excessive expression of CT-1 takes part in the myocardial hypertrophy in diabetic myocardial disease.Huangqi Injection has the inhibitory effects on the expression of CT-1 and myocardial hypertrophy.

      摘要:Objective To investigate the mechanism of Chinese herbal remedies for eliminating the tolerance of extended spectrum β-lactamases(ESBLs)-producing Escherichia coli.Methods ESBLs positive Escherichia coli were collected from clinic for establishing the drug-fast model.The sterile test tubes were divided into 12 groups and added test bacteria liquid,and the final inoculation concentration was reached to 5×105CFU/mL.The medical liquid of Qingkailing Injection(diploid concentrated solution),Shuanghuanlian Powder-Injection(freeze-dry) and cefoperazone-sulbactam sodium were prepared,added into the tubes according to test groups and two-fold diluted.The group of integration of Chinese and Western medicine was added meat soup(0.5 mL) and liquid of cefoperazone-sulbactam sodium(0.5 mL) and two-fold diluted on the 4th,6th and 8th hour respectively.After cultivated at 35 ℃ for 24 hours,the lowest concentration would be minimum inhibitory concentration(MIC) by macroscopic observation.Results The MIC of Qingkailing Injection,Shuanghuanglian Powder-Injection and cefoperazone-sulbactam sodium were respectively 23.04 g/L,62.5 mg/L and 39.06 mg/L.When Shuanghuanglian Powder-Injection was combined with cefoperazone-sulbactam sodium,the MIC of Shuanghuanglian Powder-Injection dropped by 98% and that of cefoperazone-sulbactam sodium dropped by 50%.When Qingkailing Injection was combined with cefoperazone-sulbactam sodium,the MIC of Qingkailing Injection dropped by 93.75% and that of cefoperazone-sulbactam sodium dropped by 75%.Conclusion The test proves that Qingkailing Injection,Shuanghuanglian Powder-Injection have some inhibitory effects on ESBLs-producing Escherichia coli,and when they are combined with cefoperazone-sulbactam sodium,they will have significant synergisms respectively.The sensitivity of antibiotics to drug-fast bacteria can be improved,especially when Chinese herbal remedies have effects for 4 to 6 hours.

      摘要:Objective To discuss the influences of Tangmoning Granules on the expressions of sciatic nerve growth factor(NGF) and insulin-like growth factor-1(IGF-1) proteins in rats with diabetic peripheral neuropathy(DPN).Methods Wistar rats(n=36) were randomly divided into the normal group,model group and Tangmoning Granules group.The rat model of diabetes was established by the injection of 2% streptozotocin(STZ,53 mg/kg).The diabetic rats were given orally the suspension of Tangmoning Granules(2.3 g/kg) every day during test.The sciatic ultrastructure was observed by using transmission electron microscope(TEM),and the expressions of sciatic NGF and IGF-1 proteins were detected by applying Western blot method and immunohistochemistry in all groups.Results The expressions of sciatic NGF and IGF-1 proteins were lower in the model group than those in the normal group,and those in the Tangmoning Granules group was higher than those in the model group but lower than those in the normal group.The result of TEM showed that Tangmoning Granules relieved the pathological changes of rat sciatic nerve.Conclusion Tangmoning Granules has some protective effect on sciatic nerve in diabetic rats.The mechanism may be related to the up-regulation of the expressions of NGF and IGF-1 proteins.

      摘要:Objective To establish a HPLC method for determining the plasma concentration of geniposide,arctiin and baicalin in effective fraction of Zhibangreduping Formula in rats.Methods The protein of plasma samples were precipitated with methanol-acetonitrile(1∶1) and plasma concentration was determined by using HPLC.The HPLC procedure was performed on the chromatographic column of Diamonsil C18(4.6 mm×150 mm,5 μm),and the mobile phase was acetonitrile-0.2 % phosphoric acid solution.The flow velocity was 1.0 mL/min and detection wavelengths were 238 nm and 278 nm.Results The lowest limit of detection concentrations of all constituents(S/N>3) were ranged from 0.23 mg/L to 0.32 mg/L and the average recovery was higher than 85%.RSD of intra-day and RSD of inter-day were all lower than 10%.Conclusion The method is easy and accurate,which can be used to analyse quantitatively the plasma concentration of Zhibangreduping Formula.

      摘要:Objective To study the curative effect of Jiawei Zhishixiaopi Decoction(JZD) on diabetic gastroparesis(DGP) and the mechanism.Methods All DGP patients(n=68),diagnosed as spleen deficiency and qi stagnation with stasis,and cold-heat complex,were randomly divided into 2 groups,treated respectively with JZD(treatment group,n=36) and domperidone(control group,n=32),and compared with healthy volunteers(n=30).The curative effects and changes of gastricemptying rates and gastrointestinahormone levels in two groups were observed.Results The total effective rate was higher significantly in the treatment group(90.91%) than that in the control group(70.00%,P<0.01).The gastricemptying rate was higher in the treatment group than that in the control group with a significant difference(P<0.01).The difference of levels of motilin(MOT) and gastrin(GAS) in patients of two groups was not significant compared with healthy volunteers(P>0.05) before and after the treatment.Before the treatment the difference of levels of somatostatin(SS),vasoactive intestinal peptide(VIP) and leptin(LEP) were significant in the treatment group compared with healthy volunteers(P<0.01).After the treatment the difference of levels of SS,VIP and LEP was not significant in the treatment group(P>0.05),and the difference was significant in the control group(P<0.01) compared with healthy volunteers.Conclusion JZD is an effective compound formula for treating DGP,and its curative effect may be related to down-regulation of SS,VIP and LEP levels and improvement of gastricempty in the patients.

      摘要:Objective To investigate the distribution laws of TCM syndromes of type 2 diabetic dyslipidemia,and correlation between TCM syndromes and blood-lipid changes.Methods The general materials and TCM syndromes in 60 patients with type 2 diabetic dyslipidemia were recorded and the values of total cholesterol(TC),triglyceride(TG),low-density lipoprotein cholesterol(LDL-C),and high-density lipoprotein cholesterol(HDL-C) were recorded too.Results Among 60 patients 21 with turbid phlegm obstruction syndrome,2 with spleen-kidney yang deficiency syndrome,17 with dual deficiency of qi and yin syndrome,8 with yin deficiency with yang hyperactivity syndrome,5 with qi stagnation and collateral stasis syndrome,and 7 without distinguishing syndromes.In patients with different syndrome types the values of TC,TG,LDL-C and HDL-C had no significant difference.There was a significant difference in weight index between the group of dual deficiency of qi and yin syndrome and group of turbid phlegm obstruction syndrome.There was a significant difference in the numbers of male and female patients with descending HDL-C.Conclusion In the constituent ratio of all syndrome types the proportion of turbid phlegm obstruction syndrome was the highest(35%),and that of spleen-kidney yang deficiency syndrome was the lowest(3.3%).The body index was the highest in the group of turbid phlegm obstruction syndrome and lowest in the group of dual deficiency of qi and yin syndrome.The numbers of male patients with descending HDL-C were higher than those of female patients.
