摘要:The combination proportion of Banxia ( Rhizome Pinelliae) in ZHANG Zhongjing’s formulas, measured by volume and number unit,was studied on the base of textual research on ancient and modern volume conversion standard。The method combined literature study on the species,produce places, processing with practical measurement of Banxia,and the treatment method of data statistics were applied. The results indicated that Banxia in ZHANG Zhongjing’s formulas was crude dry root of Pinellia ternata ( Thunb. ) Breit. ( family Araceae) after water washing. Half sheng of Banxia ( about 100 mL) equaled 61. 29 gram,and one piece of it was 0. 75 gram. The dosage used in ancient and modern time is different, so the dosage of Banxia should be controlled reasonably when it is used in clinic according to its combination proportion in ZHANG Zhongjing’s formulas,national pharmacopoeia and patient’s condition.

摘要:Objective To observe the effect of Naomaitong Granules combined with the mobilization of bone marrow stem cells ( BMSC) on neuron apoptosis in rats with cerebral ischemia,and investigate the possible mechanism from the influences of the combination on the expressions of relevant genes-Bax and Bcl-2,and changes of ratio of Bax to Bcl-2. Methods All rats were randomly divided into groups and the model of middle cerebral artery occlusion ( MCAO) was established by nylon thread approach. The rats of the mobilization group and combination group were injected hypodermically recombinant human granulocyte colony-stimulating factor ( rG-CSF) 3 days before the operation and 2 days after the operation. Naomaitong Granules was administrated intragastrically. The samples were taken 2,3,7 and 14 days after the operation respectively. The levels of CD34 + cells in peripheral blood and brain weredetected,the pathological changes of cerebral tissue were observed,the neuron apoptosis was examined, the expressions of Bax and Bcl-2 were detected,and the ratio of Bax to Bcl-2 was calculated. Results In the model group the levels of CD34 + cells in peripheral blood and brain increased and reached the peak on the 3rd day and 7th day respectively. In all treatment groups,and especially in the combination group, the levels of CD34 + cells increased significantly. The lesion of rat brain was significant 7 days after the ischemia,and after 14 days the lesion was alleviated in the mobilization group and combination group. In the model group the neuron apoptosis was improved,Bax expression increased,and the ratio of Bax to Bcl-2 increased on 2nd and 3rd day. In the combination group the neuron apoptosis was reduced,Bax expression decreased,Bcl-2 expression increased on 7th day and 14th day,and the ratio of Bax to Bcl-2 decreased at different time points. The comparison between the combination group and mobilization group showed that the neuron apoptosis was reduced on 2nd〗day and 3rd day,Bax expression decreased,ratio of Bax to Bcl-2 decreased,and Bcl-2 expression decreased on 7th day and 14th day at different time points in the combination group. Conclusion BMSC can be induced by cerebral ischemia entering into peripheral blood and homing in cerebral tissue,and the peak time is different. Naomaitong Granules can increase mobilized BMSC. The neuron apoptosis induced by cerebral ischemia is correlated to up-regulation of Bax expression. Naomaitong Granules can improve and prolong the anti-apoptosis effect of BMSC mobilization,which may be related to that it regulates downward Bax expression at early time ( 2nd day and 3rd day) and regulates upward Bcl-2 expression at later time ( 7th day and 14th day) .

摘要:Objective To observe the influence of Wuziyanzong Pills on the function of Sertoli cells in rats with insufficiency of kidney essence. Methods SD rats were divided into the normal group,glycosides of tripterygium wilfordii group ( GTW group) ,and high-dose,mid-dose and low-dose Wuziyanzong Pills groups ( high-dose group,mid-dose group and low-dose group) . The rat model of insufficiency of kidney essence was established by given GTW intragastrically every day. After Wuziyanzong Pills was given to relevant groups intragastrically,serum inhibin B ( INHB) ,and Johnson scores and apoptosis rate ( AR) of spermatogenic cells were detected by applying ELISA,HE staining and TUNEL. The medical serum of Wuziyanzong Pills and TM4 Sertoli cells were cultured together,and then the activity of cytochrome C oxidase ( COX) was detected by using colorimetry. Results The content of INHB and Johnson scores decreased significantly and AR increased significantly in rats with insufficiency of kidney essence ( P < 0. 05) ,while content of INHB and Johnson scores increased significantly and AR decreased significantly in the high-dose group and mid-dose group ( P < 0. 05) . The activity of COX increased significantly in the high-dose group ( P < 0. 05) . Conclusion The effect of Wuziyanzong Pills to improve the function of spermatogenic cells is related to that it can improve the function of Sertoli cells.

摘要:Objective To compare the influences of formulas with actions of soothing liver,invigorating spleen or soothing liver and invigorating spleen on thyroid function in rat model of liver-stagnation and spleen-deficiency syndrome. Method All rats were randomly divided into the normal control group ( group 1) ,model group of liver-stagnation and spleen-deficiency syndrome ( group 2) ,Chaihushugan Powder group ( group 3) ,Sijunzi Decoction group ( group 4) and Chaishusijun Decoction group ( group 5,each n = 12) . The model was established in group 2,3,4 and 5 by chronic restraint-fatigue-diet disorder method for 4 weeks. Group 3,4 and 5 were given orally the relevant formulas ( 10 mL/kg) for 2 weeks at the end of the 2nd week,and group 1 and 2 were given equivalent distilled water. On the 29th day all rats were killed and the serum levels of T3,T4,tyrotropin releasing hormone ( TRH) and thyroid stimulating hormone ( TSH) were detected. Result Compared with group 1,the serum levels of T3,T4, TRH and TSH decreased significantly in group 2 ( P <0. 05) . Compared with group 2,the serum levels of T3 and T4 increased significantly in group 3,4 and 5 ( P <0. 05) ,and the serum levels of TRH andTSH increased significantly in group 5 ( P < 0. 05) . Conclusion The hypofunction of hypothalamus-ptuitary-thyroidal axis can be observed in rat model of liver-stagnation and spleen-deficiency syndrome. Chaihushugan Powder,Sijunzi Decoction and Chaishusijun Decoction can improve thyroid function in varying degrees,and among them Chzishusijun Decoction is the best. Formulas with actions of soothing liver and invigorating spleen are related more closely to liver-stagnation and spleen-deficiency syndrome.

摘要:Objective To study the feasibility of colon-specific delivery of New Chang’ankang Formula in the treatment of ulcerative colitis ( UC) . The study was conducted through observing the influences,on rat model of acetic acid-induced UC,of gastrointestinal decomposition delivery of New Chang’ankang Formula,intragastrical delivery of the exact of Huangqi ( Radix Astragalus) ,intraintestinal delivery of berberin hydrochloride and extract of Muxiang ( Radix Aucklandiae) ,and intragastrical delivery of New Chang’ankang Formula singly. The study took New Chang’ankang Formula ( exacts of Huangqi and Muxiang,and berberin hydrochloride) as the carrier. Methods The rat model of acetic acid-induced UC was established and then the rats were divided into the normal control group,model control group, Sulfasalazine Tablets ( SASP) group,high-dose,mid-dose and low-dose groups of oral delivery,and high-dose,mid-dose and low-dose groups of colon-specific delivery. The medicinals were given to relevant groups 24 hours after the establishment of the model once a day for 6 days. The changes of fecal occult blood ( OB) were recorded every day. After the test ended,the colonic weight was weighed,the colonic length and width were measured,colonic weight index ( colonic weight/colonic length) was calculated and colonic mucosal damage index ( CMDI) and histological score ( HS) were reviewed. Results The effects of gastrointestinal decomposition delivery of New Chang’ankang Formula was as that of the same as its intragastrical delivery singly in the improvement of colonic length,weight and width,and was superior in the improvement of OB,colonic weight index,CMDI and HS. Conclusion The gastrointestinal decomposition delivery of New Chang’ankang Formula has a better protective effect on colonic mucosa in rats with acetic acid-induced UC.

摘要:Objective To observe the influences of Chinese herbal remedies ( CHR) with actions of tonifying kidney and activating blood circulation on retinal pathological changes in rat model of chronic elevated intraocular pressure ( EIOP) and investigate the mechanism. Methods The rat model of chronic EIOP was established by using cauterization of the episcleral veins,and then the rats were randomly divided into the model group,groups of high-dose,mid-dose and low-dose CHR ( treatment groups) and blank group. The rats were given CHR intragastrically for 8 weeks,and then sacrificed at the end of the 8th week. The intraocular pressure ( IOP) ,pathomorphological changes of retinal ganglion cells ( RGCs) and retinal nerve fiber layer ( RNFL) ,and apoptosis rate of RGC were observed. Results ① IOP increased significantly in rats after modeling and was 3 times higher than that before modeling after modeling for 8 weeks ( test finished) . CHR had mild effect to reduce IOP. ②The thicknesses of RNFL and RGCs layer ( RGCL) in the model group was half of those in the blank group,which was higher in all treatment groups than that in the model group,indicating that CHR could inhibit the lesion of RGCs and RNFL. ③The apoptosis rate of RGCs in the model group was 3 times higher than that in the blank group,which was lower in all treatment groups than that in the model group,indicating that CHR had significant inhibitory effect on RGCs apoptosis. Conclusion CHR with the actions of tonifying kidneyand activating blood circulation can protect the vision of rats with chronic EIOP.

摘要:Objective To study the influence of centipede acidic protein ( CAP) on myocardial apoptosis induced by angiotensin II ( AngII ) and the mechanism. Methods On the base of culturing cardiomyocytes in neonatal rats,the influence of CAP on cardiomyocyte viability was observed by using MTT method. The apoptosis was observed by using laser scanning confocal fluorescence microscope. The changes of caspase-3 activity during cardiomyocyte apoptosis were detected by using spectrophotometry with Caspase-3 Activity Assay Kit. The expression of c-fos-mRNA was detected by using RT-PCR. Results The cardiomyocyte viability decreased significantly,and cardiomyocyte apoptosis,caspase-3 activity and expression of c-fos-mRNA increased significantly in the AngII model group compared with the blank control group. The cardiomyocyte viability increased significantly,and cardiomyocyte apoptosis, caspase-3 activity and expression of c-fos-mRNA decreased significantly in the high-dose and mid-dose CAP groups compared with the AngII model group. Conclusion CAP inhibits significantly the cardiomyocyte apoptosis induced by AngII,and the mechanism is related to the decreases of caspase-3 activity and expression of c-fos-mRNA.
摘要:Objective To observe the influences of Hongbeiyegen ( Stipellaria trewioides Benth. ) on the changes of hemorheology and microcirculation in rats with renal interstitial fibrosis ( RIF) induced by unilateral ureteral obstruction ( UUO) ,and to study the mechanism of Hongbeiyegen in antagonizing RIF based on the theory of collateral disease. Methods The rat model of RIF was established by applying unilateral ureteral ligature,and then the rats were randomly divided into the model group,Hongbeiyegen group and sham-operation group. The influences of Hongbeiyegen were observed on the whole blood viscosity at shear rates of 150/s,30/s,5/s and 1/s,hematocrit ( Hct) and changes of sublingual and auricle collateral vessels. Results Hongbeiyegen reduced significantly the whole blood viscosity at shear rates of 150/s,30/s,5/s and 1/s,Hct,and levels of hyaluronic acid ( HA) ,laminin ( LN) ,type Ⅲ procollagen ( PC Ⅲ) ,type Ⅳcollagen ( CⅣ) ,serum creatinine ( SCr) and blood urea nitrogen ( BUN) index,and improved the circulation of sublingual and auricle collateral vessels. Conclusion Hongbeiyegen can improve the hemorheology and microcirculation in rats with RIF,which indicates that activating blood circulation and freeing the collaterals may be one of the mechanisms of antagonizing RIF by Hongbeiyegen.

摘要:Objective To investigate the functional status of colonic smooth muscles and relation between humoral factor and the discordance of study results in vitro and in vivo through studying the effects of Zhishi ( Fructus Aurantii Immaturus) and its medicinal serum on extracor poreal colonic muscle strips in rats with slow transit constipation ( STC) . Methods Health Wistar rats at SPF level ( n =32,half male and half female) were randomly divided into 4 groups: the normal group treated with Zhishi extract ( treatment group) ,STC group treated with Zhishi extract ( STC group) ,STC model control group ( model group) and normal control group ( normal group,each n = 8) . On the 2nd day after the modeling,the treatment group and STC group were given intragastrically the extract of Zhishi,and the model group and normal group were given normal saline. After 28 days the blood samples were collected from the abdominal aorta after giving relevant medicinals for one hour in the treatment group and STC group,and then the medicinal serum of Zhishi was separated. The serum samples collected in the normal blank group were taken as normal serum control group. The colonic muscle strips of STC rats and normalrats were incubated in a tissue chamber with Krebs liquid ( 5 mL,37 ℃) furnished with compound gas of 95% O2 and 5% CO2. The spontaneous motility of colonic muscle strips were recorded,and the influences of extracting solution and medicinal serum of Zhishi on the motility of colonic muscle strips in normal rats and STC rats. Results The extracting solution of Zhish had inhibitory effects on the amplitude and frequency of extracorporeal colonic muscle strips in normal rats,but which was not different statistically to the control value before medicine administration ( P > 0. 05) . The medicinal serum of Zhishi accelerated the amplitude and frequency of extracorporeal colonic muscle strips in normal rats,but which was not different statistically to the control value before medicine administration ( P > 0. 05) . Although the excitatory effect of the extracting solution of Zhishi on the motility of extracorporeal colonic muscle strips in STC rats was not different statistically to the control value before medicine administration ( P > 0. 05) ,the medicinal serum of Zhishi accelerated the frequency of extracorporeal colonic muscle strips ( P <0. 05) . Conclusion The extracting solution of Zhishi has different effects on extracorporeal colonic muscle strips in normal rats and STC rats. The medicinal serum of Zhishi has the excitatory effect on extracorporeal colonic muscle strips in normal rats and STC rats.

摘要:Objective To investigate the best occasion of postponing hepatic fibrosis with the therapy of reinforcing the healthy qi and eliminating toxin. Methods The rat model of hepatic fibrosis was established by the hypodermic injection of CCl4. Qilianqianhua Decoction ( QL) was given to the rats, respectively,on the first week ( QL1 group) ,forth week ( QL2 group) and seventh week ( QL3 group) after the modeling. The control group was given dimethyl diphenyl bicarboxylate ( DDB group) on the first day of the first week. The changes of levels of ATL,AST and ALB,and hepatic histopathology were observed in all groups and periods. Results After injecting CCI4,in the rats of the model group,the general condition became poorer and the body weight decreased compared with the rats of the normal group,and ascites was observed in later period. On the 6th week aminopherase increased significantly and on the 9th week serum albumin level decreased significantly. After 6 weeks obvious fibrous septum,and disorder of hepatic lobules were observed,and at the end of 9th week peudolobuli were generated extensively. The results of comparison among the OL1 group,DDB group and model group showed that the hepatic function was much better and fibrosis was to a lesser degree in the QL1 groups than that in the other groups. The results of comparison among the OL1 group,OL2 group and OL3 group showed that the hepatic function was better in the OL1 group than that in the OL2 group and OL3 group,and the aboveobserving indexes were better in the OL2 group than those in the OL3 group. Conclusion Qilianxianhua Decoction,with the actions of reinforcing the healthy qi and eliminating toxin,can postpone the development of hepatic fibrosis in rat model of hepatic fibrosis induced by CCl4. The earlier the therapy is used to intervene hepatic fibrosis,the better the effect is.

摘要:Objective To observe the anti-mutagenesis effect of extract of Tengligen ( Radix Actinidiae Chinensis) on chromosome DNA in mice,and provide the experimental base in animals for the attenuation of Tengligen during chemotherapy. Method All Kunming mice were randomly divided into groups,and the mutagenesis of DNA was induced with cyclophosphane ( CP) . The mice were given low-dose and high-dose Tengligen extract intragastrically for 4 days. The anti-mutagenesis effect of Tengligen was studied by observing the genesis of micronucleus ( MN) of polychromatic erythrocyte ( PCE) . Result The extract of Tengligen in low dose had a significant inhibitory effect on the genesis of MN of PCE. The genesis rate of MN in the low-dose group was significantly different to that in the model group ( P < 0. 05) . Conclusion The extract of Tengligen has a preventive effect on DNA lesion induced by CP.
摘要:Objcetive To set up a quantitative method for calculating multi-index constituents according to the benchmark peak of HPLC fingerprint chromatogram of effective fraction of Jiangzhining with actions of lipid regulation and anti-oxidation. Methods The HPLC fingerprint chromatogram of effective fraction of Jiangzhining with actions of lipid regulation and anti-oxidation was set up taking 2,3,5,4’-tetrahydroxy-stilbene-2-O-β-D-glucoside as the reference,and its chromatographic peak was taken as a benchmark peak to calculate the relative content of main chromatographic peaks. Results There were 10 peaks selected as the multi-constituent quantitative indexes. The total content of 10 constituents was computed with 2,3,5,4’-tetrahydroxy-stilbene-2-O-β-D-glucoside in 3 batches of effective fraction of Jiangzhining and the results showed as 12. 40% ,12. 44% and 11. 85% ,respectively. Conclusion The method of quantitative determination of multi-index constituents is easy and feasible,which can be used as one of methods in the quality control and criterion of the effective fraction of Jiangzhining with actions of lipid regulation and anti-oxidation.
摘要:Objective To select the best technique and parameter of the separation and purification of the total flavonoids from Baihuasheshecao ( Herba Hedyotis Diffusa) with macroporous resin. Method The types of macroporous resins,concentration of sample solution,pH values,volume of sample,and concentration, quantity and flow velocity of eluant were reviewed by using the single factor investigation taking the content and recovery of the total flavonoids from Baihuasheshecao as the investigative indexes. Result The parameter of the best purification technique was as follows: 0. 2 g/mL sample solution was prepared according to the radio of sample volume ( 0.4 g/mL,crude medicinal volume to resin volume) ,and pH value was 5. 0. The sample solution was rested for one hour in the chromatographic column of AB-8,eluted with deionized water of 8BV at a flow velocity of 3BV/h,and then eluted again with 70% alcohol of 5BV at a flow velocity of 2BV/ h. The content of the total flavonoids was 38. 9% in the obrained fine product,and the transfer rate was as high as 80%. Conclusion AB-8 macroporous resin has a better purifying effect on the total flavonoids from Baihuasheshecao,which is suitable for large scale industrial production.
摘要:The papers published on the Journal of Beijing University of Traditional Chinese Medicine from 2004 to 2009 were summed up. The academic distribution of TCM theoretical and experimental researches, situation of clinical reports,proportion and geographic distribution of papers from inside and outside our university,published status of papers at different project levels,sequence of the total citation frequency and impact factor ( IF) ,and quotation were analysed statistically. The purpose is to reflect the academic standard of papers on the Journal of Beijing University of Traditional Chinese Medicine and the quality of the journal,and to improve the construction and development of the journal.
