摘要:Chinese medicinal chemistry,as an important part of TCM scientific system,has some changes with the development of modern Chinese medicine.The research of Chinese medicinal chemistry involves three aspects,including nature chemistry of Chinese medicinal aiming at illustrating nature chemistry properties of Chinese medicinal and guiding new medicinal development,preparative chemistry of Chinese medicinal aiming at promoting levels of innovation and quality control and illustrating processing principle of Chinese medicinal,and pharmaceutical chemistry of Chinese medicinal aiming at drug-like effective substances,drug-like dynamic property and synergy feature.On the solid foundation of previous study,the effective substances should be taken as core,multiple disciplines should be integrated and study of system network pharmacy should be conducted in the development of Chinese medicinal chemistry.The purpose is to elucidate the scientific connotation and improve the quality of Chinese medicinal,and realize the safe,effective and quality controllable of Chinese medicinal to serve for human health well.

摘要:Traditional Chinese medicine is the most creative field of Chinese nation’s with original innovations,and it has unique and self-organized original thinking.The mode of TCM original thinking is a pioneering,particular,distinctive and creative way of thinking in traditional Chinese medicine for recognizing nature life phenomenon and solving medical problems.The paper suggested for the first time that the mode of TCM original thinking is a holism thinking mode with connotation of adopting symbols and destiny,integrating physique and mind,and unifying qi.The author also expounded the features,limitation and direction of TCM original thinking from the reason,methods and connotation construction.This study is the basic precondition of innovation of Chinese academy and culture,which shows a thinking picture of people,nature and body’s own holism and reflects the vital force of Chinese philosophy.

摘要:How did ancient TCM physicians understand water in body through the cognition to water in nature was discussed in the paper from the physical and chemical nature of water in nature perceived by human.It can be said that the knowledge about water in body and water family recognized by TCM theory is on the basis of water in nature.Human’s experience on the mobility,wettability,dissolubility and usages of water are successfully and metaphorically applied in the explanation of human physiological function,pathological changes and treatment.The theory of TCM is a kind of metaphorical true.The paper tried to prove that there is a metaphor cognition system of TCM concepts taking water as source domain in TCM theory,and a rudiment has been sketched out.

摘要:Objective To investigate the protective mechanism of fuzi polysaccharides(FPS)postconditioning to cardiomyocytes with hypoxia-reoxygenation(H/R)taking signal transducers and activators of transcription 3(STAT3)as entry point.Methods The model of cardiomyocytes with H/R was established in neonate rats and then divided into normal group,H/R group,FPS group,Stattic(specific blocker of STAT3)and FPS group(Stattic+FPS group)and Stattic group.The apoptosis rate of myocardiocytes was detected by using flow cytometer,expressions of phosphorylation STAT3(P-STAT3),cytochrome C(CytC)and caspase-3 were detected by applying Western blotting and expressions of Bcl-xl mRNA and Bcl-xs mRNA were detected by using fluosescence quantitative PCR.Results FPS postconditioning improved P-STAT3 expression,up-regulated Bcl-xl mRNA expression and down-regulated Bcl-xs mRNA expression,blocked the release of CytC and caspase-3 expression,and inhibited myocardiocyte apoptosis.Stattic can reverse the protective effect of FPS to cardiomyocytes and increase of P-STAT3 expression.Conclusion The protective effect of FPS to cardiomyocytes with H/R is related to the activation of STAT3,increase of Bcl-xl expression,protection of mitochondria,and blockage of mitochondria pathway of apoptosis.

摘要:Objective To investigate the influences of Chinese medicinal with actions of replenishing qi and nourishing yin,and resolving mass and freeing collateral vessels on transforming growth factor β1(TGF-β1) and connective tissue growth factor(CTGF) secreted by mesangial cells(MC) in rats fostered by high glucose combining Ang II,and provide a foundation for further study on prevention and treatment of diabetic nephropathy(DN).Methods The fifth generation of primary cultured MC was stimulated with high glucose and high glucose combing Ang II as well as Chinese medicinal serum.After 48 hours,the levels of TGF-β1,fibronectin(FN) and laminin(LN) were detected by using ELISA,and expression of CTGF was detected by using Western blotting method.Results Compared with control group,the levels of TGF-β1,FN and LN and CTGF expression increased significantly in other groups.Compared with high glucose+AngII group,the levels of TGF-β1,FN and LN and CTGF expression decreased significantly in irbesartan group,Chinese medicinal serum group(P<0.01).Conclusion Chinese medicinal with actions of replenishing qi and nourishing yin,and resolving mass and freeing collateral vessels can effectively decrease the level of TGF-β1,inhibit the composition of extracellular matrix and protect kidney through inhibiting CTGF expression and reducing TGF-β1 level.

摘要:Objective To study the influence of therapy of resolving accumulation and dissipating binds on expression of integrin-linked kinase(ILK) induced by transforming growth factor β1(TGF-β1) in human renal tubular epithelial cells,and expound its intervening effect in renal interstitial fibrosis(RIF).Methods The compound formula of whole formula with actions of resolving accumulation and dissipating binds was split to tonifying formula and dispersing formula.The medicinal serum samples of whole formula,tonifying formula,dispersing formula and benazepril hydrochloride were prepared by applying serum pharmacologic method.Human renal tubular epithelial cells were cultured in vitro and divided into blank group,TGF-β1 group,benazepril group,whole formula group,tonifying formula group and dispersing formula group.The different serum samples(5%) were added into renal tubular epithelial cells,and then all groups,except blank group,were added TGF-β1(10 ng/mL).After 48 hours,the total protein and total RNA were extracted.The expressions of ILK protein and its mRNA were detected by using immune Western blotting method and reverse transcription-polymerase chain reaction(RT-PCR).Results The expressions of ILK and its mRNA were less in blank group and increased significantly in TGF-β1 group(P<0.01 compared with blank group).The expressions of ILK and its mRNA decreased significantly in benazepril group,whole formula group and dispersing formula group compared with TGF-β1 group(P<0.01).The difference between tonifying formula group and benazepril group was not significant(P>0.05).Conclusion The medicinal serum of whole formula with actions of resolving accumulation and dissipating binds can inhibit the expressions of ILK protein and its mRNA,and one of the mechanisms of preventing RIF may be related to that it can intervene of signal pathway of TGF-β1/ILK.

摘要:Objective To explore the distribution mode of complex network of four-examination information in patients with coronary heart disease(CHD),and provide a new method and new thinking for study on clinical syndrome classification.Methods Taking 70 items of four-examination information as data origin from 1 480 CHD patients diagnosed by coronary angiography(CAG).The correlations among 70 nodes were set up by using mutual information method.The visible graphs of complex network of four-examination information were drawn by using Pajek 2.0 software,including different types and sizes of node graphs and k-kernel network diagram.All multi-attribute indexes were calculated for reviewing network and mining the significance of distribution mode of four-examination information.Results Among 70 items,there were 47 as nodes taking parting in the formation of network,and other 23 as isolated points.Lassitude-fatigue,forgetfulness and soreness of waist and knees with higher degree value formed the center of network,and combinational network rounded by radial nodes reflecting qi deficiency,yin deficiency,blood stasis,phlegm turbidity,yang deficiency,heat accumulation,qi stagnation and spleen deficiency.The "neighbors" of the nodes with cluster coefficient as 1 made up the groups of four-examination information and they might be the specific reflections of some syndrome types.The result of k-kernel network diagram showed that 5-kernel network of four-examination information covered mainly five syndromes including qi deficiency,qi stagnation,yin deficiency,yang deficiency and blood stasis.Conclusion The analysis technique of complex network provides an effective method for study on the distribution mode of complex network of four-examination information,which has a certain application foreground.

摘要:Objective To observe the liver injury in KKAy mice with type 2 diabetes,and study the protective effects of Huangqi(Radix Astragali seu Hedysari) and puerarin on liver.Methods Male KKAy mice were randomly divided into model group,Huangqi group and Huangqi combining puerarin group(combining group).C57BL/J mice with the same age were selected as normal group.The change of fasting blood glucose(FBG) was detected.The levels of triglyceride(TG),cholesterol(CH),alanine aminotransferase(ALT) and aspartate aminotransferase(AST) were detected after mice were sacrificed respectively when they were 24 weeks old and 28 weeks old.The pathological sections of liver tissue were prepared and stained with HE staining and oil red O staining.Results In model group,the levels of FBG,blood fat,ALT and AST increased significantly than those in normal group,and there was steatosis in liver.The size of liver was larger by naked eye in model group than that in normal group and greasy by touch.There were empty lipocytes observed by microscope in different sizes in liver cells.In Huangqi group and combining group,all biochemical indexes were improved,especially the levels of AST and ALT decreased significantly,and pathological sections showed that liver structure was complete.Conclusion There are obvious liver injury and abnormal blood fat in KKAy mice,and Huangqi or Huangqi combining puerarin can effectively inhibit abnormal blood fat and protect the structure and function of liver.

摘要:Objective To observe the influences of medicinal serum of Shudi(Radix Rehmanniae Praeparata) and Biejia(Carapax Trionycis) on the proliferation and function of osteoblasts in rats.Methods SD rats were randomly divided into 3 groups including high-dose medicinal group(high-dose group),low dose medicinal group(low-dose group) and blank group.High-dose group and low-dose group were intragastrically given decoction of Shudi and Biejia(3.250 g/kg and 1.625 g/kg) respectively twice a day and blank group was given equivalent distilled water.The medicinal serum samples of Shudi and Biejia and blank control serum samples were prepared after 3 days.The rat osteoblasts were collected after primary culturing to third generation and identifying with alkaline phosphatase(ALP).All osteoblasts were divided into 3 groups after counting and plating and treated for 72 hours.The proliferation rate of osteoblasts were detected by using MTT test,secretion of ALP was detected by using ELISA and corrected with corresponding OD value,and mRNA expression of core-binding factor α1(Cbfα1)was detected by using RT-PCR in all groups.Results The proliferation rate of osteoblasts,concentration of ALP and expression of Cbfαl mRNA were all higher in low-dose group than those in other two groups.Conclusion Shudi and Biejia can protect bone through promoting the proliferation proficiency of osteoblasts
摘要:Objective To study the chemical constituents of Emeijue Guanzhong(Lunathyrium acrostichoides Ching) for elucidating its material foundation of medical actions.Methods Emeijue Guanzhong was extracted with 95% ethanol,Abstracted with organic solvent,and isolated and purified with repeat column chromatography of silica gel and recrystallization.The obtained compounds were identified in structure according to their physicochemical features and wave spectrum characteristics.Results There were 4 compounds isolated and identified for the first time from the ethanol extract of Emeijue Guanzhong,including desmethoxymatteucinol(1),3′-hydroxy-matteucinol(2),Stigmast-4-en-3,6-dione(3),ponasterone A(4).Conclusion Compounds 1,2,3 and 4 are all isolated for the first time from Emeijue Guanzhong.

摘要:Objective To optimize macroporous resin for isolating and purifying the iridoid constituents from Zhizhuxiang(Jatamans Valeriana Rhizome).Methods The content of 11-ethoxyviburtinal was determined by using HPLC and then taking which as index,the sample solution concentration,elution solvent and elution flow rate were studied and optimized by applying single factor observation and orthogonal test design.Results AB-8 macroporous resin had excellent adsorption and desorption properties.The technological parameters were determined as that sample concentration was 0.2 g(crude Zhizhuxiang)/mL,ratio of column diameter to height was 1:6,purification solvent was 30% ethanol(2 BV),elution solvent was 90% ethanol,elution volume was 5 BV and elution flow rate was 4 BV/h.Conclusion The method was easy in operation,which provides a foundation for isolating and purifying Zhizhuxiang in large scale production.

摘要:Objective To observe the Influences of comprehensive therapy of replenishing qi,nourishing yin and activating blood circulation on urinary albumin excretion rate(UAER) and middle-term and long-term curative effects in patients with diabetic nephropathy(DN).Methods All DN patients(n=120) were randomly divided into treatment group(n=60) and control group(n=60).The control group was given present therapy and treatment group,decoction of Chinese medicinal with actions of replenishing qi,nourishing yin and activating blood circulation besides present therapy.One therapeutic course was two months and the patients were followed up on the 6th month.The level of UAER was detected before and after treatment and during follow-up on the 6th months.The curative effect on UAER and comprehensive curative effect on UAER and symptoms were reviewed 2 months after treatment and during follow-up on the 6th months.The difference in middle-term and long-term curative effects was compared between two groups 2 months after treatment.Results In treatment group UAER decreased significantly(P<0.01).The total effective rate was 68.42%,and 52.63% after 6-month follow-up,which had significant and statistical difference compared with control group(P<0.01).The review of comprehensive curative effect showed that the total effective rate was 54.39% in treatment group two months after treatment,and 52.63% after 6-month follow-up,which had significant and statistical difference compared with control group(P<0.01).Conclusion The comprehensive therapy of replenishing qi,nourishing yin and activating blood circulation can significantly decrease UAER,in DN patients.The curative effect of comprehensive therapy is better than that of present therapy and its middle-term and long-term curative effects are stable.

摘要:Objective To explore the characteristics of injured lung function in different stages and degrees of ulcerative colitis(UC),and guide clinical practice based on TCM theory.Methods The patients without diseases of respiratory system accorded with UC diagnosis were selected from outpatient department and wards of third-grade class-A hospitals in Beijing,Shanghai,Henan and Jiangsu from Sept.2009 to March,2011.The general material,symptoms,tongue and pulse manifestations and relevant risk factors of UC were investigated and at the same time,blood test and the examinations of lung function,chest X-ray,colonoscopy and histopathology were given to the patients.Results The symptom survey in this study included 261 patients.Of UC patients,there were 58.6% with chest symptoms,39.9% with short breath,28.4% with cough.The lung function survey in this stndy included 199 patients.Of 199 patients,there were 63.3% with the changes of lung function,and among which,53.3% in active stage and 53.8% with mild or moderate impairment of lung function.The types of the impairment were mainly airflow limitation(54.8%) and diffusing capacity decrease(51.6%).Conclusion The most of patients will have chest symptoms,abnormal lung function and obvious short breath and cough.The impairments of lung function manifest as airflow limitation,decrease of diffusing capacity and increase of RV/TLC%,which commonly are related to activity and severity of UC.The changes of lung function are specific in active stage and in patients with mild or moderate impairment of lung function.
