摘要:The dosage of commonly used Chinese herbs in clinical practice has changed greatly in 2000years since the pre-qin dynasty period,and it is an important issue about the research on medicinal doseeffect relationship in TCM formulas. It should be the first thing to describe the changes of dosage of commonly used Chinese herbs in clinical practice in 2000 years so as to clarify the medicinal dose-effect relationship in TCM formulas. By applying the methods of literature research and data mining,the changes of dosage of commonly used Chinese herbs in clinical practice in 2000 years since the pre-qin dynasty period were calculated and described,the concept of dosage valley of Chinese herbs in clinical practice was proposed. The dosage valley was made with historical period as the length and clinical dosage range of commonly used Chinese herb as the width,to describe the changes of dosage of commonly used Chinese herbs in clinical practice. The significance of study on clinical dosage valley of commonly used Chinese herbs is reflected in three aspects: knowing the history of TCM formulas application,providing some documentary evidences for clinical medication,and understanding the information about history,geography,humanities and society. There are six factors which affect dosage valley,including personal knowledge and experience of doctors,characteristics of monthly,yearly climate and diseases,factor of academic schools,factors of economy,politics,geography or traffic,influence of science,impact of government management and regulation,et,al. The dosage valley of some Chinese herbs represented by Gegen( Radix Puerariae),Shigao( Gypsum Fibrosum) and Fuzi( Radix Aconiti Lateralis Preparata) in 2000 years,are statistically described in details. Some issues about the safety and effectiveness of Chinese herbs in clinical practice are proposed.

摘要:In the wave of bringing science to Traditional Chinese Medicine during the period of the Republic of China,dictionaries of Chinese medicine emerged at the historic moment based on the requirements of scientific research,clinical,learning and society. In this period the dictionaries of Chinese medicine were still imperfect in compilation style,item setting and retrieval system although they elementarily met the basic requirements as a dictionary. As a carrier of Chinese pharmacy system,these dictionaries complied during the period of the Republic of China focused on scientificity and practicality in content,opposed abusing yin-yang and five elements theory so as to cast mists before one’s eyes. They had made contributions to summarize and popularize the knowledge of Chinese herbs and thus to influence on the development of Chinese pharmacy.

摘要:Xerophthalmia is a commonly-encountered disease and has gradually become a hot topic of ophthalmic research in recent years. Although local pathological changes of eyes appear in the course of xerophthalmia,systemic pathological changes of body are pathogenesis of this disease considering from holistic concept and syndrome differentiation concept of Traditional Chinese Medicine. The liver opens into the eyes and its fluid is tear. Xerophthalmia is caused by liver depression,yin deficiency,and disorder of distribution of body fluids. So the main pathogenesis of xerophthalmia rely on liver depression and yin deficiency,and the treatment on xerophthalmia with the therapy of soothing liver and nourishing yin can achieve good curative effect.

摘要:Objective To compare the influences of disassemble formulas of Yizhi Fang with different combinations on peroxidative and apoptosis of cells induced by Aβ25- 35,and to investigate the mechanism of Chinese medicinal with actions of tonifying deficiency( Buxu medicinal) and those with actions of resolving turbidity( Huazhuo medicinal) in reducing the toxicity of Aβ25- 35,as well as their reasonable combination proportion. Methods The cell model of Alzheimer’s disease( AD) was established by adding Aβ25- 35into cortical neurons of primary cultured rat. Yizhi Fang was modified into Buxu formula and Huazhuo formula according to medicinal nature. Normal group,Quanfang group,Buxu group,Huazhuo group,Bu-hua 2: 1 group and Bu-hua 1: 2 group were established and the medicated serum samples were added into corresponding groups respectively. Blank control group was not given Aβ25- 35( non-Aβ group) and model group was given Aβ25- 35( Aβ group). The neurocyte survival rate,leakage rate of lactate dehydrogenase( LDH),activity of superoxide dismutase( SOD) and malondialdehyde( MDA)content were detected and calculated by using chromatometry in all groups after 24 hours. The expression of Caspase3 was observed by using immunohistochemical method. The apoptosis of cells was observed by using TUNEL assay. Results In Aβ group( 20 μmol / L),the survival rate of neurocyte and activity of SOD decreased,and LDH leakage rate,MDA content increased,expression of Caspase3 and apoptotic cell counting increased( P < 0. 01). The medicated rat serum samples could increase the survival rate of neurocyte,decrease LDH leakage rate and MDA content,improve the expression of Caspase3,and reduce apoptotic cells( P < 0. 01 or P < 0. 05). The effectiveness was the best in Quanfang group and Buxu group( P <0.01 or P <0.05),secondly in Bu-hua 2: 1 group and Bu-hua 1: 2 group( P <0. 01 or P <0. 05),and weaker in Huazhuo group( P < 0. 01 or P < 0. 05). Conclusion Aβ25- 35is neurotoxic to cause the peroxidation injury and excessive apoptosis of neurocyte. The Buxu medicinal is better than Huazhuo medicinal in inhibiting peroxidation and apoptosis,while Huazhuo medicinal should be combined with Buxu medicinal for improving curative effect. Buxu medicinal plays a major role in inhibiting neurotoxicity of Aβ25- 35.
摘要:Objective To observe the curative effect and mechanism of TCM therapy of warming yang and invigorating spleen on constipation,and its influences on the expressions of colon c-kit protein and SCF protein in rats. Methods The rat model of constipation was established by using laxatives,and treated with the formula granules of Chinese medicinals with actions of warming yang and invigorating spleen for 4 weeks,in which the proportion of Baizhu( Rhizoma Atractylodis Macrocephalae) to Roucongrong( Herba Cistanches) was 2 to 1. The rats were divided into high-dose group( 9 g / kg),mid-dose group( 4. 5 g / kg) and low-dose group( 2. 25 g / kg),and compared with mosapride group. The quantity,weight and form of feces of rats in all groups were observed,the intestinal transport function was detected by using activated carbon suspension propulsion method,and the expressions of colon c-kit protein and SCF protein in rats were detected by using Western blot test. Results The therapy of warming yang and invigorating spleen had an obvious curative effect on rats with constipation( quantity,weight and form of feces and intestinal transport function),and showed significant differences when compared with model group. The experimental results indicated that the expressions of c-kit protein and SCF protein were significantly lower in model group than those in normal group with statistical significance,and increased in all treatment groups after treatment. The difference in the expression of c-kit protein was statistically significant between treatment groups and model group. The expressions of SCF protein increased in all treatment groups and were higher than that in model group,but the difference was not statistically significant,which might be related to the small sample size of the study.Conclusion The mechanism of laxative constipation in rats may be related to the decreased expressions of c-kit protein and SCF protein. The therapy of warming yang and invigorating spleen has an obvious curative effect on laxative constipation and can increase the expressions of c-kit protein and SCF protein.

摘要:Objective To compare the influences of Buzhong Yiqi Tang( BYT) with different compatibilities on gastrointestinal function in rats with spleen deficiency fever. Methods Male Wistar rats( weighted from 200 g to 220 g) were randomly divided into normal group,model group,BYT group,BYT without Chai-Sheng group,BYT with high-dose Chai-Sheng group and high-dose Chai-Sheng group( each n = 10). The rat model with spleen deficiency fever was established by using the methods of improper diet,over strain and injection of lipopolysaccharide( LPS). The body weight of rats in all groups was detected every week,and the changes of behavior were observed and recorded. On the 18th day all the groups were intragastrically given corresponding decoctions respectively for 5 consecutive days.On the 22th day the levels of gastrin( GAS),motilin( MTL) and somatostatin( SS) in blood were detected in rats of all groups. Results In model group,the integral of behavior and level of SS in peripheral blood increased,body weight,levels of GAS and MTL in peripheral blood and D-xylose excretion rate in urine decreased significantly compared with normal group. In BYT group,BYT without Chai-Sheng group and BYT with high-dose Chai-Sheng group,the integral of behavior and level of SS in peripheral blood decreased,body weight,levels of GAS and MTL in peripheral blood and D-xylose excretion rate in urine increased significantly compared with model group. In high-dose Chai-Sheng group,the integral of behavior and level of SS in peripheral blood increased,body weight,levels of GAS and MTL in peripheral blood and D-xylose excretion rate in urine decreased( P < 0. 05 or P < 0. 01).Conclusion Conspicuous gastrointestinal dysfunction appears in the rats with spleen deficiency with fever,which can be alleviated significantly by BYT. BYT without Chai-Sheng and BYT with high-dose Chai-Sheng have decreased curative effects,and the simple compatibility of high-dose Chai-Sheng has no curative effect. The effects of invigorating spleen and supplementing qi of BYT may be related to the dose and combination of Shengma( Rhizoma Cimicifugae) and Chaihu( Radix Bupleuri).

摘要:Objective To observe the changes of neurons and synapses in hippocampus CA1 zone in amyloid protein precursor( APPswe) / presenilin( PS1dE9) double transgenic mice( AD model),and to review the influence of curcumin on the cognitive function in APPswe / PS1dE9 double transgenic mice.Methods All APPswe /PS1dE9 double transgenic mice( 9 months old) were randomly divided into model group,rosiglitazone( 10 mg / kg / d) group,and high-dose( 400 mg / kg / d),mid-dose( 200 mg /kg /d) and low-dose( 100 mg / kg / d) curcumin groups( high-dose group,mid-dose group and low-dose group,each n = 6),and C57 / BL6J mice with the same age and genetic background were assigned into the normal control group. All groups,except normal group,were intragastrically given relevant medicinal respectively once a day for 6 months. The changes of hippocampus ultrastructure were observed with transmission electron microscope( TEM). Results The structure of hippocampus neutrons and neuropils were normal in normal group. In model group,neuron pyknosis,higher electron density,increase of lipofuscin( LPF) and lipid droplet in cytoplasm,and degeneration of neutrons in varying degrees were observed in the hippocampus CA1 zone,and myelinic degeneration,decrease and structural abnormality of synapses,and increased aggregation of synaptic vesicles were observed in neuropils. In curcumin groups the number of synapses increased and the above mentioned changes were ameliorated compared with model group. Conclusion Curcumin may improve the learning and memory abilities in APPswe /PS1dE9 double transgenic mice through increasing the number of synapses and changing hippocampus ultrastructure.
摘要:Objective To study the influence of Fuzheng Toudu Qudu Fufang combining modified Qinghao Biejia Tang on CD34+derived dendritic cells( DC) in the patients with minimal residual disease in myelogenous leukemia( MRD-L). Methods Bone marrow mononuclear cells( BMNC) were separated by using Ficoll centrifugation in the patients with MRD-L. CD34+cells were isolated by using immuno-magnetic mircobeads and cultured for amplification to get DC,which was induced by medicated serum of Chinese medicinal in different concentrations in vitro. The morphologic characteristics of DC were observed with the inverted phase contrast microscope,and the expressions of DC surface molecules,CD83,CD80,CD86、CD1a and HLA-DR,were detected by using flow cytometry. Mature DC was induced for activating T lymphocytes. The kill effect of activated T lymphocytes on K562 at different ratio of effectors to target cells was detected by using methyl thiazolyl tetrazolium test( MTT). Results CD34+was promoted by different medicated serum combined cytokines to differentiate into DC with typical morphology after being cultured for 9 days. The expressions of CD83,CD80,CD86and HLA-DR increased in high-dose + cytokines group,mid-dose + cytokines group,low-dose + cytokines group and cytokines group compared with FCS serum group and rabbit serum group( P < 0. 01). The expression of CD1a increased in mid-dose + cytokines group and low-dose + cytokines group compared with high-dose +cytokines group and cytokines group( P < 0. 01),and there were no significant difference among other groups. Cytotoxicity of T lymphocytes to K562 cells were activated by DC. The kill rates of K562 in highdose + cytokines group,mid-dose + cytokines group and low-dose + cytokines group were all higher than those in cytokines group,FCS serum group and rabbit serum group,and the kill rates in mid-dose +cytokines group and low-dose + cytokines group were higher or significantly higher than those in cytokines group,FCS serum group and rabbit serum group( P < 0. 05,P < 0. 01). Cytotoxicity of T lymphocytes went up along with up-regulation of ratio of effectors to target cells. The cytotoxicity to K562 cells of T lymphocytes derived from health people showed no significant difference compared with those derived from patients( P > 0. 05). Conclusion Modified Qinghao Biejia Tang can promote CD34+to transform into DC in the patients with MRD-L.

摘要:Objective To investigate the influence of therapy of soothing liver and fortifying spleen on visceral hypersensitivity in rats and to study the mechanism. Methods The rat model was established with maternal separation( MS). All rats were randomly divided into high-dose,mid-dose and low-dose Jiawei Xiaoyao Wan groups( high-dose group,mid-dose group and low-dose group),normal saline group( NS group),model group and normal group( half male and half female). The abdominal withdrawal reflex( AWR) was observed and graded during colorectal distension( CRD) in all groups. The expressions of corticotrophin-releasing factor( CRF),spinal cord dorsal horn neurons c-fos and colon5-HT3R were observed by using immunohistochemistry technique,and content of serum CRF was detected by using enzyme-linked immunosorbent assay( ELISA). Results ① MS improved the visceral hypersensitivity in rats especially in female rats,and reduced significantly the threshold of CRD( P <0. 05). ② In Jiawei Xiaoyao Wan groups,the difference in the threshold of CRD was not statistically significant before treatment,and threshold of CRD increased significantly after treatment( P < 0. 05).③The content of CRF in hypothalamus and serum decreased as the dose of Jiawei Xiaoyao Wan increased in Jiawei Xiaoyao Wan groups,and the difference was statistically significant between high-dose group and mid-dose group or between mid-dose group and low-dose group( P < 0. 05). ④The expressions of spinal c-fos and colon 5-HT3R decreased as the dose of Jiawei Xiaoyao Wan increased showed a tendency of the expressions being more obvious as the increase of medicinal dose( P < 0. 05). Conclusion MS can improve and Jiawei Xiaoyao Wan can decrease the visceral hypersensitivity,which has a tendency of the changes being more obvious as the increase of medicinal dose. The mechanism may be related to the inhibition of HPA axis and visceral sensitivity through reducing CRF secretion and release and inhibiting the expressions of spinal c-fos and colon 5-HT3R.

摘要:Objective To study the extraction and purification processes of alkaloids in Heye( Folium Nelumbinis),conduct further the research on pharmacological activity and pharmacokinetics of the alkaloid extracts,and lay a foundation for large-scale production and clinical application. Methods The content of total alkaloids,aporphine and nuciferine were determined by using acid-dye colorimetry and HPLC. The best extraction process was optimized by applying orthogonal design test,and the purification process with positive ion-exchange resins was investigated. Results The best extraction process was as follows: 90% ethanol( 16 times of crude Heye) for extracting for 3 times and 1. 5 hours each time,and the best purification process was as follows: 90% ethanol extractive fluid of Heye was evaporated under reduced pressure,and then the residue was given ultrasonic dispersion( 20 times,mass fraction = 1%HCI) and centrifugation for obtaining supernatant. The supernatant was eluted with 50% ethanol( 5 BV)after flowed through D001-CC positive ion-exchange macroporous resin,and eluted again with enthanol ammonia solution [7 BV,mass fraction = 1%,70% enthanol( V / V) ]at flow velocity of 10 BV / h. The enthanol ammonia eluent was collected,evaporated after reducing pressure,and the residue was dried under reduced pressure. Finally the alkaloid fractions of Heye were obtained. Conclusion The processes are simple with low cost and pollution,and the content of total alkaloids in the alkaloid fraction of Heye is over 50%.

摘要:Objective To optimize the extraction and enrichment process of phenolic acids in Danggui( Radix Angelicae Sinensis). Methods The experiment was carried out taken ferulic acid as indicator according to the condition in L9( 34) orthogonal design table for investigating the influences of ethanol concentration,solvent amount and duration and frequency of extraction on extraction results and optimizing extraction process. The enrichment process was optimized through investigating the types of macroporous resins,ratio of diameter to height,pH and concentration of sample solution,maximum volume of sample,washed volume,ethanol concentration and volume for edulcoration and elution and flow velocity. Results The final preferred extraction and enrichment process of phenolic acids in Danggui was as follows: 8 times amount of 70% ethanol,reflux extracting for 2 times,1 hour each time,combining and concentrating extracts to 0. 5 g / mL,centrifugating for 30 min( 4 500 r / min),supernatant putting in HPD-400 macroporous resins,sample amount being 4. 5 g / mL,adsorption flow velocity being0. 5 mL / min,ratio of diameter to height being 1: 7,distilled water( 3 BV) and 20% ethanol( 2 BV) for edulcoration,flow velocity of edulcoration being 1 mL / min,50% ethamol( 6 BV) for elution,and flow velocity of elution being 2 mL / min. Conclusion The content of ferulic acid is 151. 8 mg / g in concentrates after enriched with the extraction and enrichment process optimized in the paper,and the process is stable and viable.

摘要:Objective To observe the distribution characteristics of syndrome factors and disease locations in the patients with chronic heart failure( CHF) combining diabetes mellitus( DM). Methods An inventory survey method was used in 631 CHF patients,and then all patients were divided into CHF combining DM group and CHF group according to whether they had DM. The data was collected including age,sex,height,weight,syndrome factors and disease locations. All data was analyzed statistically and the distribution laws were discussed. Results There were 169 patients in CHF combining DM group and 462 in CHF group. The most patients were old and male. The average age was( 68. 78 ±9. 86) years old in CHF combining DM group and( 66. 65 ±11. 02) years old in CHF group,and average weight was( 70. 97 ± 13. 20) kg in CHF combining DM group and( 66. 61 ± 11. 62) kg in CHF group( P < 0. 05). The major syndrome factors were qi deficiency and blood stasis in CHF patients,while the distributions of water fluid and phlegm turbidity were more in CHF combining DM group than that in CHF group( P = 0. 025,P = 0. 026). The factors involving disease locations were mainly heart,kidney,spleen and lung in CHF patients,and the percentage of kidney and lung was significantly higher in CHF combining DM group than that in CHF group( P < 0. 05). Conclusion Compared with CHF patients without DM,the age and weight are higher in the patients with CHF combining DM,and the distributions of syndrome factors,such as water fluid,phlegm turbidity,kidney and lung,are more.

摘要:Objective To study the curative effect and mechanism of Bushen Huoluo Fang in treatment of polycystic ovary syndrome( PCOS,type of kidney deficiency and blood stasis). Methods The patients( n =78) with PCOS( type of kidney deficiency and blood stasis) were randomly divided into treatment group( n = 40) and control group( n = 38). The treatment group was orally given Bushen Huoluo Fang and control group was given Diane-35 for 3 months. The curative effect,levels of testosterone( T) and luteinizing hormone( LH),follicle-stimulating hormone( FSH),volume of ovary and follicles counting were observed after treatment. Results The effective rate was 97. 5% in treatment group and 78. 9% in control group( P < 0. 05). The levels of T and LH and LH / FSH decreased significantly in two groups than before treatment( P < 0. 05). The volume of ovary and follicles counting were significantly reduced in two groups,and all indexes were better in treatment group than those in control group( P < 0. 05).Conclusion The curative effect of Bushen Huoluo Fang is better than that of Diane-35 in treatment of PCOS( type of kidney deficiency and blood stasis),and the mechanism may be related to decreasing levels of T and LH and regulating endocrine disorders.

摘要:Objective To observe the influences of TCM syndrome differentiation and treatment on anxiety and depression status in the patients with functional dyspepsia( FD). Methods The study applied randomized,double-blind,placebo-controlled and multi-center clinical trials. All FD patients( n= 273) were divided into treatment groups and control groups respectively with damp-heat congesting syndrome,spleen deficiency and qi stagnation syndrome,liver-stomach qi stagnation syndrome and depression-heat in liver and stomach syndrome according to TCM syndrome differentiation and treatment,and then treated with corresponding formula granules or simulant of Chinese medicinal. The changes of integrals of self-rating anxiety scale( SAS),self-rating depression scale( SDS) and symptoms were observed before and after treatment. Results The percentage of anxiety patients was significantly higher in group of liver-stomach qi stagnation syndrome than that in group of spleen deficiency and qi stagnation syndrome( P < 0. 05). The integrals of SAS,SDS and symptoms decreased significantly in treatment groups and control groups after treatment( P < 0. 05),and decreased more significant in treatment groups than those in control groups( P < 0. 05). The integrals of SAS and SDS in treatment groups with all syndromes,and symptom integrals in control groups and treatment groups with all syndromes decreased significantly after treatment( P < 0. 05). The integrals of SAS and SDS decreased significantly in control group of liver- stomach qi stagnation syndrome( P < 0. 05),and were significantly higher in treatment group of spleen deficiency and qi stagnation syndrome than those in control group( P < 0. 05).Conclusion TCM syndrome differentiation and treatment can relieve the status of anxiety and depression at the same time of relieving symptoms in FD patients. Xiangsha Liujunzi Tang combining modified Zhizhu Wan has significant curative effect on anxiety and depression in FD patients with spleen deficiency and qi stagnation syndrome.

摘要:Objective To provide clinical data for the diagnosis and treatment of chronic heart failure( CHF) through investigating the epidemiological features in the patients with CHF due to coronary heart disease( CHD) in Chinese medicine hospitals. Methods The epidemiological questionnaire of CHF was formulated for the survey in the patients with CHF due to CHD in 17 Chinese medicine hospitals.Results There were totally 439 patients chosen,and most of them were elderly. The percentage of male to female was 1. 3∶ 1. 0 and average age of the male was lower than that of the female. The complicated diseases were mainly hypertension, diabetes, arrhythmia and cerebrovascular diseases. The major incentives were overwork,aggravated original diseases,mood swing and infection. The syndrome factors involving disease nature with higher frequency were orderly qi deficiency( 81. 5%),blood stasis( 79. 5%),water fluid( 27. 3%),phlegm turbidity( 20. 0%),yin deficiency( 19. 1%) and yang deficiency( 18. 2%). The syndrome factors involving disease locations were mainly heart( 90. 4%),secondly lung( 33. 0%),spleen( 32. 6%),kidney( 31. 7%) and liver( 15. 7%). In the distribution of syndromes,the common syndromes were syndrome of qi deficiency and blood stasis and syndrome of dual deficiency of qi and yin. Conclusion The distribution of syndromes in CHF patients shows commonly co-occurrence of deficient and excessive syndromes, and the study provides some epidemiological evidences for TCM diagnosis and treatment in the patients with CHF due to CHD.
