摘要:The pulse-time relationship model in Neijing established under the guide of the theory of unity of heaven and man can be divided into 3 types,that is yin-yang pulse-time relationship,five-phase pulsetime relationship and 3-yin 3-yang pulse-time relationship.The five-phase pulse-time relationship is used the most commonly in clinic,and 3-yin 3-yang pulse-time relationship is rarely mentioned.Neijing attached importance to the grasp of pulse-time relationship based on the idea of correspondance between man and universe.

摘要:Collateral vessels of viscera and bowels refers to deep ones in meridian system related closely to the characteristics of viscera and bowels,and they have their own features and are one important part of collateral vessel system.The paper initially investigated deep collateral vessels of viscera and bowels based on the theory of collateral invaded by chronic disease brought up by YE Tian-shi and ancient literature.The purpose is to analyze the function,distribution,examination,characteristics of syndromes and treatment,and relationship among 15 collateral vessels from the aspect of standardizing TCM terms.

摘要:Objective To understand whether Xiaodu Shuan can inhibit Treg cell functin to change the local immune microenvironment for inhibiting tumor growth through detecting the protein expressions of cytotoxic T lymphocyte antigen-4(CTLA-4) and programed death-1(PD-1).Methods Treg cells and CTL were separated by using immunomagnetic beads.The protein expressions of CTLA-4 and PD-1 were detected by using Wstern-blot method.Results In CTL-Treg co-culture group,the protein expressions of CTLA-4 and PD-1 increased significandy,which decrased significantly after the intervention ofβ-elemene and Xiaodu Shuan especially in Xiaodu Shuan group.Conclusion Xiaodu Shuan andβ-elemene all can inhibit the up-ragulation effect of Treg on CTLA-4 and PD-1 in CTL.

摘要:Objective To investigate the melacular mechanism of Xinjia Guishen Wan(a compund formular with the actions of tonifying kidney and activating blood) in promoting follicular development taken the proliferation of granulocytes as an entry point.Methods The model of follicular development disorder was established by using GTW in rats,and then the rats were treated with high-dose,mid-dose and low-dose Xinjia Guishen Wan for 2 weeks.The ovary index was calculated,and granulocyte thickness was detected with light microscope after HE staining.The expressions of ovary CyclinD2 mRNA and granulocyte CyclinD2 protein were detected respectively by using Taqman RT-PCR and immunohistochemistry method.Results The ovary index,granulocyte thickness and expressions of ovary CyclinD2 mRNA and granulocyte CyclinD2 protein all decreased in model group compared with normal group(P <0.05 or P <0.01).After treatment with Xinjia Guishen Wan in different doses,the abovementioned indexes all increased especially in high-dose group and mid-dose group(P < 0.05 or P <0.01).Conclusion Xinjia Guishen Wan can accelerate the progress of cell cycle,promote granulocyte proliferation,and further improve follicular development through up-regulate the expression of CyclinD2.

摘要:Objective To discuss the influences of curcumin on carbohydrate metabolism and differentiation of preadipocytes and mechnism.Methods The model of adipocyte insulin-resistant was established by using culturing-inducing combination method of high-dose glucose and insulin.The uptake of 3H-glucose was detected in adipocytes after inventioned with curcumin,and the differentiation degree of adipocytes was analyzed by using spectrophotography quantitatively after oil red 0 staining.The expressions of PPAR-y and C/EBPa were detected by using real-time fluorescence quantitative polymerase chain reaction(RT-PCR).Results The uptake rate of glucose was 134.51%in curcumin group compared with control group,and the differentiation of adipocytes and expressions of PPAR-y mRNA and C/EBPa mRNA were improved significantly compared with control group(P <0.05).Conclusion Curcumin can improve glucose uptake and differentiation of adipocytes and expressions of PPARγ mRNA and C/EBPa mRNA.

摘要:Objective To reveal the laws of human intestinal tract adapting seasonal and climate changes and provide invidence for TCM theory of correspondence between nature and human through observing different levels of interleukin-2(IL-2)in rat jejunum,ileum and colon in five seasons of Chinese medicine.Methods Pinealectomized rat model was taken as study object,and the changes of IL-2levels in rat jejunum,ileum and colon in five seasons(spring,summer,latter summer,autumn and winter)and changes after pinealectomizing were observed.Results In normal group,the level of IL-2was higher in jejunum in latter summer compared with other 4 seasons(P <001),higher in latter summer and winter in ileum compared with spring,summer and autumn(P <0.01 or P <0.05)and higher in colon in winter compared with other 4 seasons(P<0.01).After pinealectomizing,the normal seasonal and climate rhythm of IL-2 secretion was disorder in all intestinal sections.Conclusion The level of IL-2 has seasonal and climate changes in rat jejunum,ileum and colon,which is consistent with TCM theory of correspondence between nature and human.The pineal gland takes part of the rhythmic regulation of intestinal function.

摘要:Objective To study the influences of Jiedu Tongluo Injection on the activities of cerebral superoxide dismutase(SOD),glutathione peroxidease(GSH-PX),catalase(CAT) and malondialdehyde(MDA) in rats with middle cerebral artery occlusion(MCAO).Methods The model of MCAO was established in rats by using Koizumi and Nagasawa methods.The cerebral samples were collected and homogenized after cerebral ischemia for 2 h and reperfusion for 24 h.The influences of Jiedu Tongluo Injection on the activities of MDA,SOD,GSH-PX and CAT were observed and detected by using biochemistry methods.Results The activities of SOD,CAT and GSH-PX were promoted and MDA content was reduced in high-dose,mid-dose and low-dose Jiedu Tongluo Injection groups.Conclusion Jiedu Tongluo Injection has preventive effect on free radical injury after cerebral ischemia.

摘要:Objective To investigate the influence of Yiqi Huatan Huoxeu Fang on inflammatory axis of thyroid interlukin-23(IL-23)/interlukin-17(IL-17)in NOD.H-2h4mice.Methods Female NOD.H-2h4 mice(8 weeks old) were chosen and divided into normal group,model group,low-dose Yiqi Huatan Huoxeu Fang group(low-dose group,20.8 g/kg) and high-dose Yiqi Huatan Huoxeu Fang group(high-dose group,62.4 g/kg).Normal group was given double distilled water and other groups,Nal water(0.05%).After 8 weeks,the model of autoimmune thyroiditis(AIT)was established and intervened for 8 weeks.The levels of serum total T3(TT3)and total T4(TT4)were detected by using radioimmunoassay(RIA),and level of serum thyroid stimulating hormone(TSH)was detected by using solid-phase chemi-luminescent enzyme immunoassay.The morphological changes of thyroid cells were observed with light microscope.The protein expressions of thyroid mesenchymal CD68,and thyroid IL-17and IL-23 were detected by using immunohistochemistry technique,mRNA expressions of IL-17 and IL-23 were detected by using RT-PCR,and percentage of splenocyte Th17 was analyzed by using flow cytometry.Results The levels of TT3,TT4 and TSH had no significant difference compared among all groups(P >0.05).The incidence of AIT,expressions of CD68,IL-17 and IL-23 and percentage of Th17 all increased significandy in model group compared with normal group(P <0.01).The expressions of CD68,IL-17 and IL-23 decreased significandy(P < 0.01) and percentage of Th17 decreased(P < 0.01 or P <0.05) in high-dose group and low-dose group compared with model group.Conclusion The mechanism of Yiqi Huatan Huoxeu Fang for treating AIT may be related to that it can inhibit the inflammatory axis of IL-23/IL-17 and down-regulate percentage of Th17.
摘要:Objective To investigate the effect of Compound Buguzhi Liniment on vitiligo.Methods The model of vitiligo was established by using chemical decolorization and then treated with Compound Buguzhi liniment in different doses.Compound Kaliziran Tincture was taken as a positive control drug.The influences of Compound Buguzhi Liniment on distribution of skin melanin and activities of cetylcholinesterase and tyrosinase were observed.Results Compound Buguzhi liniment promoted skin pigmentation.The level of melanin increased significantly in basal layer cells and spinous layer cells,and hair follicles.The activities of cetylcholinesterase and tyrosinase were improved significantly.Conclusion Compound Buguzhi Liniment has a preventive and therapeutic effect on experimental vitiligo.
摘要:Objective To observe the influences of Guipi Tang on phenotype Sca-1 and CD34+ of hematopoietic stem cell(HSC) and cell cycle in benzolism mice,and discuss its possible mechanism in hematopoietic regulation.Methods Kunming mice(n =72,8-12 weeks old,half male and half female) were randomly divided into normal group,model group,Western drug group,and high-dose,mid-dose and low-dose groups(each n = 12).The benzolism model was established by putting mice into a manual simulated condition with high concentration benzene(2000 mg/m3) for 8 h a day for 14 d continuously,until the level of WBC dropped below 4.5 × 109/L.The high-dose,mid-dose and low-dose groups were orally given solution of Guipi Tang(0.072g/mL,0.036g/mL and 0.018g/mL) respectively and normal group and model group were orally given normal saline water(20 mL/kg) once a day for 3weeks.The influences of Guipi Tang on mitotic cycle in peripheral blood and bone marrow and phenotype Sca-1 and CD34+ of HSC were detected by using automotive blood cell analyzer and flow cytometry.Results HSC was activated,expressions of Sca-1 and CD34+ increased and primordial HSC increased in mid-dose group and Western drug group(P <0.05).In high-dose group,the cell transition from G0/G1 phase to S phase and from S phase to G2/M phase were promoted,proliferation index increased significantly(P <0.05 or P <0.01),and levels of WBC and HBG increased.The change of platelet was not significant.Conclusion Guipi Tang may improve the quantity and function of early HSC and hemopoietic ability through promoting the cell transition from G0/G1 phase to S phase and from S phase to G2/M phase.
摘要:Objective To establish a HPLC method for simultaneous determination of 7 kinds of organic acid in Acanthopanax sessiliflorus(chlorogenic acid,neochlorogenic acid,caffeic acid,1,3-dicaffeoylquinic acid,3,4-dicaffeoylquinic acid,3,5-dicaffeoylquinic acid and 4,5-dicaffeoylquinic acid).Methods The HPLC procedure was performed on the chromatographic column of Hypersil 0DS2 C18(250×4.6 mm,5 μm) at room temperature,and mobile phase was 0.1%formic acid in acetonitrile-0.1%formic acid in water.The flow velocity was 1.0 ml/min and detection wavelength was 324 nm.Results All 7 kinds of organic acid showed good linear relationship respectively at range of 20.5-328 μg/mL,1.99-31.8 μg/mL,2.43-38.9 μg/mL,2.44-39.0 μg/mL,2.52-40.3 μg/mL,7.25-116 mg/L and 2.42-38.7 μg/mL.The average recoveries of them were within a range from 95.2%to 103.9%.Conclusion The method is quick,simple,accurate and sensitive,which can be used as a method for controlling the quality of Acanthopanax sessUiflorus and other Acanthopanax materials.

摘要:Objective To establish a analysis method of LC-MS/MS to study the internal change features of norcantharidin(NCTD) in high dose,medium dose and low dose.Methods The HPLC procedure was performed on the chromatographic column of CAPCELL PAK MG II C18 at room temperature.The mobile phase was acetonitrile-water(containing 0.1%formic acid) in gradient elution,and flow velocity was 0.2 mL/min.A tandem mass spectrometer equipped with electrospray ionization(ESI) source was used for detecting in the mode of multiple reaction monitoring(MRM,ion pair m/z 169.3→123.1 for NCTD and ion pair m/z 267.1→135.1 for internal standard ribavirin).The concentration of NCTD was detected in different rat tissues by using LC-MS/MS.Results The linear range of NCTD was(0.01-10.00)μg/mL in heart,liver and lung,and(0.10- 10.00) μg/mL in spleen and kidney.Intra-day RSD and inter-day RSD were less than 14%in all groups and extraction recovery was between 81.0%and 106%.After intravenous injection of NCTD in 3 dose levels,the dynamic changes in all tissues(except of spleen) were similar.The concentration of NCTD reached to the peak after one hour and then rapidly decreased.The distribution level of NCTD was higher in liver and kidney showing a dosedependent manner.Conclusion The LC-MS/MS method is simple,sensitive and accurate,which can be used for study the distribution of NCTD in rat tissues.When dose of NCTD is higher,its concentration is also higher in kidney,which may be related to its renal toxicity.The study will provide a reference for rational administration of NCTD in clinic.

摘要:Objective To study the best technology conditions of smashing extraction method for extracting baicalin from Huangqin(Radix Scutellariae).Methods The influence factors on the smashing extraction method were investigated by using orthogonal design taking the extraction rate of baicalin as an index.The extractive rate,extraction time and electricity consumption were compared with other methods reported in titerature.Results The best technique of the smashing extraction method for extracting baicalin from Huangqin was as follows;Huangqin was immersed in boiling water(100℃) for 10 min for killing enzyme and retaining baicalin.After filtration,the soak solution was added 65%ethanol(20 times)and given smashing extraction for 2 min,and then merged with extractive solution.Conclusion The smashing extraction method for extracting baicalin from Huangqin is easy and economic with higher extractive rate compared with decocting method and reflux method,which can be taken as a reference for utilization of Huangqin.

摘要:Objective To investigate the distribution laws of TCM syndrome types of advanced stomach cancer and provide evidences for clinical treatment.Methods According to relative diagnostic standard,6 basic syndrome types commonly in clinic were chosen,and 167 patients were given syndrome differentiation,classification and statistical analysis.Results The distribution frequency of TCM syndrome types were,in a descending order,syndrome of spleen-stomach deficiency cold,syndrome of qi stagnation and blood stasis,syndrome of dual deficiency of qi and blood,syndrome of stomach-heat damaging yin,syndrome of liver qi invading stomach,and syndrome of phlegm-damp congealing.Conclusion Syndrome of spleen-stomach deficiency cold is the most common syndrome types in the patients with advanced stomach cancer.

摘要:Objective To discuss the correlation between Kupperman scores and TCM constitutional types in menopausal women.Methods A TCM constitution questionnaire was used for investigating TCM constitutional types in 130 menopausal women and the results were analyzed.Results ①Age,BMI,menarche age,marriage age,primiparous age,pregnant times and puerperal times had no statistical significance in menopausal women with different Kupperman scores(P > 0.05).②Kupperman scores were negatively correlated to score of normal constitution,and positively correlated to scores of other 8types of abnormal constitution(P < 0.01).There was significant difference among menopausal women with the same constitutional types but different Kupperman scores(P < 0.01),and score of normal constitution decreased as menopausal symptoms exacerbated.③The menopausal symptoms were mosdy mild and medium in women with normal constitution,and severe in women with other abnormal constitutions.Conclusion Constitutional types is closely correlated to degree of menopausal symptoms,and differentiation of constitutional types will be helpful to prevention and treatment of menopause syndrome in clinic.
摘要:Objective To observe the difference in the expressions of FAH gene and PTGS2 gene regulated by acupoint selection on the same meridian in patients with migraine.Methods The patients(n = 20) were diagnosed as migraine(type of hyperactivity of liver-yang),and then divided into acupoint group and non-acupoint group(each n-10).The acupoint group was given needling in Jiaosun(TE20),Waiguan(TE15),Yanglingquan(GB34) and Qiuxu(GB40),and non-acupoint group was given needling in 4 non-acupoints once a day for 10 times.The samples of peripheral vein blood were collected from all patients before and after treatment,and genetic expression profiles were obtained by using gene chip technology.After Gene Ontology(GO) analysis,the profiles were reviewed with Visual Analogue Scale(VAS) before and after treatment in two groups.Results The expressions of FAH gene and PTGS2 gene decreased significantly in all samples in acupoint group(FAH:P <0.05,PTGS2:P <0.05 after GO analysis),while there were no expressions of these two genes observed in non-acupoint group.The results of VAS review showed that there was no difference between two groups before treatment(P>0.05).In acupoint group,there was difference before and after treatment(P <0.05),and there was difference after treatment and after follow-up(P < 0.05).In non-acupoint group,there was difference before and after treatment(P < 0.05),and there was no difference after treatment and after follow-up(P >0.05).The comparison in D-value in two groups before and after treatment showed difference(P <0.05).The immediate curative effect and long-term curative effect were more significant in acupoint group than those in non-acupoint group.Conclusion Acupoint selection on the same meridian has curative effect on migraine without aura(type of hyperactivity of liver-yang) and one of mechanisms may related to regulating FAH gene and PTGS2 gene.

摘要:Objective To observe the influences of acupuncture on the protein expressions of myocardial CFTR and ClC-2(regulatory proteins of chloride channel) in rats with acute myocardial ischemia(AMI),and discuss the mechanism of specificity of selecting acupoints along meridians.Methods SD rats(n =60) were randomly divided into blank group,model group,Neiguan(PC6) group,Lieque(LU7) group and non-acupoint group.Except of blank group,other groups were given multiple subcutaneous injection of isoproterenol for establishing AMI model.The Neiguan group,Lieque group and non-acupoint group were given electro-acupuncture in bilateral Neiguan,Lieque and middle point between Tianshu(ST25) and Shenque(CV8) once a day for 7 d.The changes of myocardial histology were observed by using microscope after HE staining.The content of creative kinase(CK-MB) was detected by using kit method,content of malonyldialdehyde(MDA) was detected by using EELISA,and protein expressions of CFTR and C1C-2 were detected by using Western blot method.Results The HE staining sections of myocardial tissue showed that fragmentation and disordered arrangement of myocardial fibers,sarcoplasm swelling,obvious inflammatory cell infiltration,and wide distribution of many brown granules in nucleus in model group,Lieque group and non-acupoint group,which were relieved in Neiguan group compared with model group.Compared with blank group,the protein expressions of CFTR and C1C-2 increased significantly(P < 0.01),CK-MB content increased significantly(P < 0.01) and MDA content increased in model group,Lieque group and non-acupoint group.Compared with model group,CK-MB content decreased(P<0.05),MDA content decreased(P <0.05) and expressions of CFTR and C1C-2 decreased significantly(P < 0.01) in Neiguan group.Conclusion Acupuncture in Neiguan can effectively relieve the pathological changes induced by myocardial ischemia,and the mechanism may be related to regulating the protein expressions of CFTR and C1C-2.
