摘要:ObjectiveTo study the expression of pituitary key functional genes and their pituitary-specific transcription factor 1 (Pou1fl) gene in H22 tumor-bearing mice with different TCM patterns.Methods190 Kunming mice were injected subcutaneously into the armpit with H22 hepatoma carcinoma cells, and 60 more healthy mice were assigned into the normal group. The mice with pattern of exuberance of pathogenic toxin, asthenia of qi, asthenia of yang and qi, or asthenia of qi, yin and yang were identified by using the method of mice standardization of four diagnosis information and syndrome differentiation developed by our team. Then RNA was extracted from pituitary gland, hypothalamus and testis and was detected using GeneChip Mouse Exon 1.0 ST Array. And the genetic data of pituitary gland of different patterns were analyzed.ResultsCompared with normal group, the expression of pituitary key functional genes including Pomc, Gh and Prl were decreased by about 10% in mice with pattern of exuberance of pathogenic toxin or asthenia of qi, which was the pattern of early stage. At the middle or late stage, TCM patters were the pattern of asthenia of yang and qi, or asthenia of qi, yin and yang; Pomc, Gh, Prl and Tshb were upregulated by more than 30%. Pou1fl expressed at the highest level in pituitary tissue with the characteristic of tissue specific expression. Pou1f1 and Neurod1 were only upregulated in pattern of exuberance of pathogenic toxin, Tbx19 and Egr1 were upregulated in all patterns, which was just opposite to that of Nr4a1, Nr4a2, Pitx1, Gata2, Nr4a3, and Foxl2. Poulfl target genes including Crem, Tcf4, Rnf41, Lrpprc, Mrps18b, Eif3a, Lars, Arf4, Cep57, Vcpip1, Ptpn6, Myd88, F3, Lrrfip3 and Dixdc1 all expressed in the similar pattern with that of Poulfl, upregulated in pattern of exuberance of pathogenic toxin.ConclusionThe abnormal expression of pituitary key functional genes was proved in the tumor-bearing mice with different TCM patterns. Pituitary-specific transcription factor 1 with its target genes expressed in the similar way in the pattern of exuberance of pathogenic toxin.
摘要:Disease-syndrome is a frequently used term in clinical Chinese medicine. Disease-syndromes are body’s abnormal states affected by pathogenic factors with the same basic pathology,which are classified under the guidance of traditional Chinese medicine theory. Basic pathology is essential for the classification of disease-syndromes. It is the internal stable connection among all elements in diseasesyndrome,and reflects and runs through the whole progress of disease-syndrome. In addition,basic pathology is also the internal basis used for determining the main symptoms of disease-syndromes and the laws for diagnosis and treatment in Chinese medicine. The implication of disease-syndrome is different from disease,syndrome and symptom. To properly understand the implication of disease-syndrome has a contribution to summarizing the previous clinical experience and guiding the clinical practice.
摘要:Hernia’s name was recorded as early as in Hanshu Yiwenzhi,and also in Huangdi Neijing,there are nearly 20 articles about the disease. Because the discussions were very messy,they have resulted in different understandings,diagnoses and therapies to hernia in later generations. However,if carefully combing the original text of Huangdi Neijing,the characteristics of hernia can be found as follows: the locations of hernia are commonly in the liver meridian and kidney meridian. There are two main pathogeneses of hernia,one is qi derangement resulting in internal stagnation,and another is cold congealing. Therefore the therapeutic methods for hernia should include replenishing fire origin,and regulating and tonifying liver and kidney,or soothing liver and regulating qi,or warming the interior and dissipating cold,and moving qi and eliminating dampness.
摘要:Objective To study the expressions of dynorphin( Dyn) and prodynorphin( PDyn) in cortex and striatum in rats with Tourette syndrome( TS),and interventional effect of Jianpi Zhidong Tang( JZT) on them. Methods Male SD rats( n = 60) were randomly divided into model group( n = 45)and control group( n = 15),and the model group was given intraperitoneal injection of IDPN for establishing TS model. After successful modeling,all rats were taken in the test and randomly divided into model group,Tiapride group and JZT group( each n = 15). The model group and control group were given orally normal saline water( 0. 9%),and other 2 groups were given,respectively,Tiapride( 21mg/kg) and JZT solution( 16 g/kg) for 6 w. The integrals of TS rat stereotyped behaviors were recorded after the last drug delivery at every week. The expressions of Dyn and PDyn in cortex and striatum were detected, respectively, by using enzyme-linked immunosorbent assay( ELISA) and real-time fluorescence quantitative polymerase chain reaction( RT-PCR). Results(1) The integrals of TS rat stereotyped behaviors showed a descending tend in Tiapride group and JZT group,and the last time observation showed that the integrals were lower in Tiapride group and JZT group than those in model group( P < 0. 05),and the integrals were lower in JZT group than those in Tiapride group( P < 0. 05).(2)The content of cortex Dyn and relative expression of PDyn mRNA had no significant difference among all groups( P > 0. 05). The content of striatum Dyn and relative expression of PDyn mRNA increased significantly in model group compared with control group( P < 0. 05),were lower in Tiapride group and JZT group than those in model group( P < 0. 05),and had no significant difference compared with control group( P > 0. 05). Conclusion The higher expressions of striatum Dyn and PDyn may take part in TS rat stereotyped behaviors. The anti-tics mechanism of JZT may be related to inhibiting higher expression of striatum Dyn and reducing facilitatory effect in direct pathway.
摘要:Objective To observe the protective effect of Rooibos( endemic plant in the South Africa) in mice with cerebral ischemia-reperfusion injury( IRI). Methods All mice were divided into control group,IRI group,low-dose Rooibos group( low-dose group,2. 8 g/kg),mid-dose Rooibos group( middose group,5. 6 g/kg) and high-dose Rooibos group( high-dose group,11. 2 g/kg). Before modeling,3 medicated groups were orally given Rooibos once a day for 7 d,and control group and IRI group were given isometric distilled water. After one week,the model was established by using cerebral middle cerebral artery occlusion( MCAO),and then all groups were continuously given Rooibos or distilled water for 7 d. The influence of Rooibos on cerebral infarction volume was observed after TTC staining.The influence of Rooibos on movement,cognition and sense was observed after neurologic score test,balance beam test,Morris water maze( MWM) test and corner test. The influence of Rooibos on redox state after cerebral injury was observed after detecting the activities of superoxide dismutase( SOD) and glutathione peroxidease( GSH-PX),and content of malondialdehyde( MDA). The influence of Rooibos on inflammatory state after cerebral injury was observed after detecting the brain contents of interleukin-1β( IL-1β),tumor necrosis factor-α( TNF-α) and intercellular adhesion molecule-1( ICAM-1).Results The cerebral infarction volume decreased significantly( P < 0. 05),scores of balance beam test decreased significantly( P < 0. 01),times from corner to non-injury side decreased significantly( P <0. 01),and contents of TNF-α,IL-1β and ICAM-1 decreased significantly( P < 0. 01 or P < 0. 05) in 3Rooibos groups compared with IRI group. Conclusion Rooibos has certain protective effect in mice with cerebral IRI,and the effect may be related to the decrease of inflammation factors in brain tissue.
摘要:Objective To observe the influence of medicated serum of Yiqi Huatan Sanyu Fang on apoptosis of pulmonary artery smooth muscle cells( PASMCs) in hypoxia rats, and discuss the relationship between its mechanism and mitochondrial pathway. Methods The medicated serum of Yiqi Huatan Sanyu Fang and blank serum were prepared. PASMCs were cultured by using tissue explant method and given identification by using a-SMA immunofluorescence. After the best medicated serum and best hypoxia time were screened by using methyl thiazolyl tetrazolium test( MTT),PASMCs were divided into normoxia control group,hypoxia model group,5% medicated serum group( low-dose group),10%medicated serum group( mid-dose group) and 20% medicated serum group( high-dose group,medicated serum diluted by blank serum). The effective duration was,respectively,12 h,24 h and 48 h. PASMCs were divided,according to MTT results,into normoxia control group,hypoxia model group and best medicated serum group in formal test. The apoptosis of PASMCs was detected by using TUNEL-FITC/DAPI,and expressions of casepase-9,casepase-3,Bcl-2 and Bax protein were detected by using Western Blot in all groups after the effect on the best hypoxia time by medicated serum. Results The results of MTT showed that when the concentration of the medicated serum was 20% and hypoxia time was 24 h,the inhibitory effect was the highest with the best result. The results of TUNEL-FITC/DAPI showed that the medicated serum improved significantly the apoptosis of PASMCs compared with hypoxia model group( P < 0. 01) under hypoxia condition. The results of Western Blot showed that the medicated serum promoted the expressions of casepase-9,casepase-3 and Bax protein( P < 0. 05,P < 0. 01),and reduced expression of Bcl-2 protein( P < 0. 01). Conclusion The medicated serum of Yiqi Huatan Sanyu Fang can induce the apoptosis of PASMCs in hypoxia rats maybe through mitochondrial pathway.
摘要:Objective To observe the influence of surrounding-hoop method( Hongzhongxiao Tincture)under guidance of defended field theory on expressions of plasma high mobility group box-1 protein( HMGB1) and lipoxin A4( LXA4) in rats with skin abscess. Methods All rats( n = 40) were randomly divided into blank group,model group,control group,pretreatment group and treatment group.Except of blank group,other 4 groups were given hypodermic injection of 1 mL Staphylococcus aureus( 3-5 × 10109CFU/mL) for establishing the model of skin abscess. The model group was given normal saline dressing,control group was given Mupirocin Ointment dressing,pretreatment group was given 2mL Hongzhongxiao Tincture dressing 4 h after injection of Staphylococcus aureus,and treatment group was given 2 mL Hongzhongxiao Tincture dressing over 1 cm around abscess once a day. The blood samples( 2 mL) were collected respectively after 3 d,7 d,14 d and 18 d. The expressions of HMGB1 and LXA4 were detected by using enzyme-linked immunosorbent assay( ELISA). Results The content of HMGB1 was lower in model group compared with blank group( P < 0. 01),and higher in pretreatment group and treatment group compared with model group( P < 0. 01 or P < 0. 05) 3 d after treatment. The content of LXA4 increased in model group,control group and treatment group compared with blank group( P < 0. 01 or P < 0. 05) 3 d after treatment. The content of LXA4 was lower in pretreatment group than that in model group( P < 0. 01). The content of LXA4 increased in control group and blank group compared with model group( P < 0. 05) 18 d after treatment. Conclusion Early surrounding-hoop method( Hongzhongxiao Tincture) can improve the expression of plasma LXA4 through taking part in early inflammation inhibition,and shortening inflammatory progress at the same time,promoting HMGB1 release at early treatment stage and inhibiting its release and/or stopping its proinflammatory activity at middle and late treatment stages for improving effectively the formation of defended field.

摘要:Objective To observe the protective effect of Kudiezi Injection( KI) on vascular endothelial cells,and study initially the effective mechanism of PKCδ/MARCKS signal pathway after hypoxia injury.Methods The hypoxia injury model was induced by CoCl2 in human umbilical vein endothelial cells( HUVEC),and then divided into normal group,model group,KI group and Rottlerin group. The cytomorphologic changes were observed by using light microscope 24 h after hypoxia injury,vitality of HUVEC was detected by using methyl thiazolyl tetrazolium test( MTT),and distributions and expressions of p-PKCδ and p-MARCKS were detected by using immunocytochemical method and Western Blot in all groups. Results KI and Rottlerin alleviated significantly the cytomorphologic changes,and enhanced significantly the vitality of cells after hypoxia injury( P < 0. 01). The expressions of p-PKCδ and pMARCKS were significantly increased in model group compared with normal group( P < 0. 05),and were significantly inhibited by KI and Rottlerin( P < 0. 05). Conclusion KI has a protective effect on vascular endothelial cells after hypoxia injury,and the mechanism is related to down-regulation of vitality of PKCδ/MARCKS signal pathway.
摘要:Objective To discuss the influence of Tongluo Baoshen Fufang( TBF) and its disassembled formulas on P-IRE1α, promoter of endoplasmic reticulum stress( ERS) and GRP78, molecular chaperone, in diabetic rats with hyperglycemic kidney injury induced by streptozocin( STZ).Methods Male SD rats were randomly divided into normal group and modeling group. The model of diabetes was established by applying intraperitoneal injection of STZ in model group. After successfully modeling,the modeling group was randomly divided into model group,valsartan group( 8. 93 mg/kg),TBF group( 19. 43 g/kg),Tongluo Baoshen Buxu group( TBB group,6. 92 g/kg) and Tongluo Baoshen Jiedu group( TBJ group,12. 5 g/kg). The general conditions of rat daily activities were observed in all groups. The expressions of p-IRE1α and GRP78 were analyzed semi-quantitatively by using immunohistochemistry and IPP software after 6 weeks and 16 weeks. Results P-IRE1α and GRP78 mainly existed in the cytoplasm of proximal and distal renal tubule cells and glomerular cells observed by light microscope. The results of semi-quantitative immunohistochemistry and IPP software showed that the expressions of P-IRE1α and GRP78 increased in model group compared with normal group after 6 weeks and 16 weeks. Compared with model group,the expressions of P-IRE1α and GRP78 decreased in valsartan group,TBF group,TBB group and TBJ group( P < 0. 05) after 6 weeks and 16 weeks. The comparison among valsartan group,TBF group,TBB group and TBJ group did not show difference( P >0. 05). Conclusion The rat model of kidney injury induced by hyperglycemia has ERS activation. TBF and its disassembled formulas can take play a renal protective role through inhibiting ERS. It can be speculated that the central pathogenesis of diabetic hyperglycemic kidney injury is deficiency of the root and excess of the tip in Chinese medicine.
摘要:Objective To investigate the myocardial protective effect of Yiqi Yangyin Huoxue Fang( YYHF) in rats with type 2 diabetes mellitus( T2DM). Methods Male Wistar rats( aged 12 weeks old,n = 60) were chosen and divided randomly into blank group( n = 10) and modeling group( n =50). The model of T2DM was induced by high-fat diet and low-dose streptozocin( STZ). After successfully modeling, the modeling group were randomly divided into groups and given orally,respectively,normal saline water( model group),Fufang Danshen Diwan( FDD group,70 mg/kg),and high-dose,mid-dose and low-dose YYHF( high-dose group,mid-dose group,low-dose group,55. 00 g/kg,27. 50 g/kg,13. 75 g/kg). once a day for 4 weeks. The routine biochemical indexes including blood sugar and blood fat were detected. The myocardial tissue was collected from the anterior left ventricular wall,and then a part of which was prepared to slices for observing myocardial cytomorphologic changes after HE stain by using light microscope,and other part of which was taken for detecting content of nuclear factor of activated T cell 4( NFAT-4) by using Western Blot,and expression of miR-133 a by using real-time fluorescence quantitative polymerase chain reaction( RT-PCR). Results The levels of blood sugar,glycated serum protein( GSP) and blood fat increased significantly in model group compared with blank group( P < 0. 05),and decreased significantly in all treatment groups compared with model group( P < 0. 05). The content of NFAT-4 increased significantly( P < 0. 05),and expression of miR-133 a decreased significantly( P < 0. 05) in model group compared with blank group. The expression of miR-133 a increased significantly( P < 0. 05 or P < 0. 01),and content of NFAT-4 decreased significantly( P < 0. 05 or P < 0. 01) in all treatment groups compared with model group. YYHF and FDD alleviated myocardial cytomorphologic changes under light microscope. Conclusion YYHF can significantly ameliorate glycolipid metabolism,inhibit cardiomyocyte hypertrophy and protect myocardial cells,which may be related to the regulation of expressions of miR-133 a and NFAT-4.

摘要:Objective To provide the evidence for prevention and treatment of aspirin resistance( AR) in coronary artery disease( CAD) patients by analyzing the prevalence,related risk factors of AR and the distribution rules of TCM patterns. Methods One hundred CAD patients were eligible,who were confirmed by angiography and took aspirin at dose of 100 mg per night for at least four weeks,at the average age of( 68. 95 ± 8. 91) years. All the patients were divided into three groups according to the platelet aggregation assay induced by arachidonic acid( AA,1 mmol/L). The platelet aggregation rate of≥80%,≥50% and < 80%,or ≤50% was used to define AR,aspirin semiresistance( ASR) or aspirin sensitive( AS) respectively. Meanwhile,based on the experts advice and literature retrieving,TCM pattern factors of patients were identified as qi deficiency, sputum turbid, blood stasis, qi stagnation,yang deficiency,yin deficiency and cold stagnation. Then the risk factors and distribution of TCM patterns were compared among the three groups. Results There were 12 CAD patients of AR found,whose fasting blood glucose levels,serum creatinine,quantity of lesion and severe lesion in the coronary artery were higher than those of other two groups( P < 0. 05),but insulin levels two hours after meal was lower( P < 0. 05). There were no significant difference of TCM pattern distribution among the three groups( P > 0. 05). And the multivariate logistic regression analysis showed that serum fasting glucose level was a significant risk factor of aspirin resistance. Conclusion AR has a relative high prevelance. Fasting blood glucose level is closely associated with AR in CAD patients. In AR group,the deficiency pattern factors included qi deficiency and yin deficiency,while the main excessive pattern factors were blood stasis and sputum turbid. Aspirin sensibility has no effects on distribution of TCM pattern factors.

摘要:Objective To observe the clinical efficacy of Huazhuo Jiedu Fang in patients with chronic erosive gastritis( CEG,syndrome of turbidity-toxin accumulating in the interior),and investigate its effect mechanism based on the expression changes of protein kinase B( Akt) and hepatocyte growth factor receptor( c-Met) in gastric mucous tissue and serum before and after treatment. Methods The patients( n = 80) were randomly divided into 2 groups according to the design of randomized single blind controlled clinical trials,and treatment group was treated with Huazhuo Jiedu Fang and control group was treated with Alatan Wuwei Wan. The expression levels of Akt and c-Met in serum and gastric mucous tissue were observed in 2 groups before and after treatment. Results The expression level of Akt decreased significantly in treatment group compared with control group( P < 0. 05) in serum and gastric mucous tissue,and expression level of c-Met in gastric mucous tissue decreased significantly in treatment group compared with control group( P < 0. 05). Conclusion Huazhuo Jiedu Fang can reduce significantly expression levels of Akt and c-Met in serum and gastric mucous tissue in patients with chronic erosive gastritis CEG( syndrome of turbidity-toxin accumulating in the interior). The effect mechanism may be that Huazhuo Jiedu Fang can regulate expression levels and activities of Akt and c-Met.

摘要:Objective To investigate the protective effect of acupuncture combining madopar on dopaminergic neurons and possible mechanism through observation on the influence of acupuncture in chorea-tremble controlled zone combining madopar on cerebral protein kinase B( Akt) expressions in mice with Parkinson ’s disease( PD). Methods Male C57BL/6 mice were randomly divided into normal group,model group,madopar group,acupuncture group( A group) and acupuncture combining madopar group( A + M group). The mouse model of PD was established by intraperitoneal injection of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine( MPTP),and madopar group,A group and A + M group were given different therapies. All mice survived for 28 d after modeling. The ethnology of mice was observed by using pole climbing method. The lost of nigra dopaminergic neurons and Akt expressions were detected by using immunohistochemistry technique. Results The time of pole climbing were significantly shorter in madopar group,A group and A + M group than that in model group( P < 0. 05).After modeling,the expression of nigra dopaminergic neurons decreased in madopar group,A group and A + M group than that in normal group( P < 0. 05),and increased in A + M group than that in model group and madopar group( P < 0. 05). The expression of Akt in nigra compacta increased significantly in A + M group than that in other groups( P < 0. 05). The expressions of Akt at zones of CA1,CA3 and CA4 in hippocampal part increased significantly in A + M group than those in model group and madopar group( P < 0. 05),and the expressions at zones of CA1 and CA4 increased significantly than those in A group( P < 0. 05). Conclusion Acupuncture in chorea-tremble controlled zone,especially combining madopar then,can get a therapeutic effect on PD through possibly regulating PI3K/Akt pathway.
摘要:Objective To observe the influence of electro-acupuncture( EA) on expression of endometrium phosphorylation insulin-like growth factor-1 receptor( p-IGF-1R),and signal transduction pathways of MAPK and PI3 K in mice with controlled ovarian hyperstimulation( COH),and discuss the mechanism of EA improving embryo implantation. Methods Female Kunming mice were randomly divided into natural cycle group,COH group,EA group,EA + BMS-536924 group,EA + LY-294002 group and EA + PD-98059 group( each n = 20),and mice were divide again into donor mice and recipient mice. The model was established by applying controlled hyperstimulation program,and given EA in Guanyuan( RN3),Zhongji( RN4) and Sanyinjiao( SP6) at the day of injecting human chorionic gonadotropin( HCG) once a day until stopping collecting samples. The fertilized ovums were collected from donor mice and cultured in vitro,and then transplanted into the uterus of recipient mice. The clinical pregnancy rate and site number of embryo implantation were observed. The expressions of endometrial IGF-1R,p-IGF-1R,phosphorylation protein kinase( p-Akt) and phosphorylation extracellular regulated protein kinases1/2( p-ERK1/2) were detected by using Western Blot. Results The clinical pregnancy rate,site number of embryo implantation and expressions of endometrial IGF-1R,p-IGF-1R,p-Akt and p-ERK1/2 were all lower in COH group than those in natural cycle group( P < 0. 05 or P < 0. 01). All indexes were higher in EA group,EA + LY-294002 group and EA + PD-98059 group than those in COH group( P < 0. 05 or P < 0. 01). But the clinical pregnancy rate,site number of embryo implantation had no significant improvement in EA + BMS-536924 group,and expression of p-IGF-1R was significantly lower than that in natural cycle group( P < 0. 01). The comparison among EA + BMS-536924 group,EA + LY-294002 group and EA + PD-98059 group showed that the expression of p-Akt was higher in EA + PD-98059 group, and expression of p-ERK1/2 was higher in EA + LY-294002 group( P < 0. 01).Conclusion Embryo implantation can be improved by EA in COH mice,which may be related to EA can up-regulate the expression of p-IGF-1R and signal transduction pathways of PI3K/Akt and MAPK/ERK1/2.

摘要:Objective To explore whether or not 3 Xi-cleft points in 3 yin meridians of foot can specifically respond primary dysmenorrheal through observing body surface resistance manifestations of Xicleft points in Spleen Meridian( SP),Kidney Meridian( KI) and Liver Meridian( LR) related closely to uterus. Methods Healthy female university students( normal group,n = 78) and those with primary dysmenorrheal( dysmenorrheal group,n = 78) were chosen from the Beijing University of Chinese Medicine and the Dongzhimen Hospital. The acupoints-Diji( SP8),Zhongdu( LR6) and Shuiquan( KI5) were chosen for monitoring continuously and dynamically body surface resistance in 2 groups for30 min on the 1std of menstruation and 3 d after menstruation finished. The resistance imbalance degree( RID) at the same acupoint was taken as outcome indicators. Results Finally there were 48 students in normal group and 46 in dysmenorrheal group after screening according to inclusion,exclusion and abortion criteria. The between-group and within-group comparison in RID of 3 Xi-cleft points at different time points showed that RID at Diji( SP8) had significant difference between the 1std of menstruation and 3 d after menstruation finished( P = 0. 04) in dysmenorrheal group. The between-group and withingroup comparison in RID of Zhongdu( LR6) and Shuiquan( KI5) had no significant difference between 2 groups( P > 0. 05) at different time points. Conclusion Diji( SP8) is initially speculated as a possible responding acupoint for primary dysmenorrheal.

摘要:Objective To illuminate the central mechanism of analgesic effect of Huaisanzhen on nerve root pain of the fifth lumbar vertebra( L5) of lumbar disc herniation( LDH) by applying resting functional magnetic resonance imaging( fM RI). Methods The patients( n = 10) with L5 nerve root pain of LDH were given the first resting fM RI, then acupuncture in acupoint of Gentong 2 of Huaisanzhen,and finally the second resting fM RI. The obtained test imagines and data were analyzed and processed by using relevant software. Results The brain regions activated by pure resting state included bilateral paracentral lobules,cingulate gyrus,thalamus and occipital lobe,third periventricular zones,optic chiasm,midbrain periaqueductal gray and cerebellar vermis and other parts. The brain regions activated by acupuncture included bilateral occipital lobe,temporal lobe,parietal lobe and left side of the midbrain,and some scattered activation regions in cerebellum. The brain regions activated after acupuncture included bilateral temporal lobe,left thalamus,splenium of corpus callosum,optic chiasm,right internal capsule,pons,midbrain periaqueductal gray and other parts. Conclusion L5 nerve root pain causes the changes of brain function areas,and activities and active levels of resting state default network are inhibited and reduced. After acupuncture in acupoint of Gentong 2,the signals reaches the functional regions of brain through nerve pathways and they were activated or inhibited resulting in analgesic effect.
